摘 要:试验旨在研究槲皮苷(QUE)对正常3D4/2细胞吞噬活性及对感染猪圆环病毒Ⅱ型(PCV2)的3D4/2细胞吞噬活性的影响。试验设置细胞对照组、0.05%DMSO对照组和不同浓度的槲皮苷药物QUE组(12.5、25.0、50.0、100.0、200.0、400.0μmol/L),采用中性红法测定QUE在36 h内对3D4/2细胞吞噬活性及对感染PCV2的3D4/2细胞吞噬活性的影响。结果表明,与对照组相比,25.0μmol/L和50.0μmol/L的QUE处理后显著提高了3D4/2细胞的吞噬指数(P<0.05);100.0~400.0μmol/L的QUE处理后极显著提高3D4/2细胞的吞噬指数(P<0.01)。与PCV2阳性对照组相比,不同浓度的QUE (12.5~400.0μmol/L)处理36 h后均能够极显著提高PCV2感染3D4/2细胞的吞噬指数(P<0.01)。与对照组相比,100.0、200.0、400.0μmol/L的QUE极显著提高了细胞吞噬指数(P<0.01)。研究表明,QUE对3D4/2细胞具有一定的保护作用与免疫调节作用,对其吞噬活性具有良好的促进作用。The study was carried out to investigate the effect of quercetin(QUE) on phagocytic activity of 3D4/2 cells and the phagocytic activity of 3D4/2 cells infected with PCV2. In the experiment, cell control group, 0.05% DMSO group and QUE drug group in the concentrations of 12.5, 25.0, 50.0, 100.0, 200.0 or 400.0 μmol/L were set up, and the effect of quercetin on 3D4/2 cell phagocytosis activity and the PCV2 infected 3D4/2 cell phagocytic activity were determined by neutral red method. The results showed that compared with control group, QUE treatments of 25.0 μmol/L and 50.0 μmol/L significantly increased the phagocytosis index of 3D4/2 cells(P<0.05). The phagocytosis index of 3D4/2 cells was extremely increased after QUE treatment with 100.0~400.0 μmol/L(P<0.01). Compared with PCV2 positive control group, QUE at different concentrations(12.5~400.0 μmol/L) could extremely increased the phagocytosis index of PCV2-infected 3D4/2 cells after 36 h treatment(P<0.01). Compared with control group, QUE of 100.0, 200.0 and 400.0 μmol/L extremely increased the phagocytosis index(P<0.01). The study indicates that QUE has certain protective and immunomodulatory effects on 3D4/2 cells, and has a good promotion effect on its phagocytosis activity.
