摘 要:试验旨在探究IGF-1基因单核苷酸突变位点对保山猪和杜洛克猪生长性能的影响。试验以保山猪和杜洛克猪为研究对象,采用直接测序法对IGF-1基因的多态性进行检测,利用生物信息学软件分析编码蛋白结构。结果显示:在保山猪IGF-1基因编码蛋白的外显子上共筛选出1个SNP位点(Exon4-G189A),该突变属于同义突变,引起密码子改变,但所编码的氨基酸未改变。生物信息学分析结果显示,保山猪IGF-1基因CDS区编码164个氨基酸,分子式为C782H1265N247O239S11,相对分子质量为18303.82,理论等电点为9.89,不稳定性指数为73.25,整体表现为亲水性,不具有跨膜结构,含有信号肽序列;蛋白质二级结构只含有无规则卷曲和α-螺旋,分别占60.37%、39.63%,三级结构预测模型与保山猪IGF-1基因的蛋白结构匹配度为-1.15,具有较好的一致性。研究表明,IGF-1基因在物种进化过程中高度保守,使其编码保山猪和杜洛克猪的氨基酸序列相同,蛋白质结构和功能差异很小或不存在差异,能否作为调控保山猪瘦肉少、脂肪多、生长慢的候选基因还需进行关联性的研究。The purpose of the experiment was to explore the effect of IGF-1 gene single nucleotide mutation on growth performance of Baoshan pigs and Duroc pigs. In the experiment, Baoshan pig and Duroc pig were selected as the research objects. The polymorphism of IGF-1 gene was detected by direct sequencing, and the structure of coding protein was analyzed by bioinformatics software. The results showed that a total of one SNP site(Exon4-G189A) was screened on the exon of the protein encoded by the IGF-1 gene of Baoshan pig. The mutation was a synonymous mutation that caused the change of the codon, but the encoded amino acid did not change. The results of bioinformatics analysis showed that the CDS region of Baoshan pig IGF-1 gene encoded 164 amino acids, the molecular formula was C782H1265N247O239S11,the relative molecular weight was 18 303.82, the theoretical isoelectric point was 9.89, the instability index was 73.25, and the overall performance was hydrophilic, without transmembrane structure, and contained signal peptide sequence. The secondary structure of protein only contains irregular curls and α-Helix, accounting for 60.37% and 39.63% respectively.The matching degree of the tertiary structure prediction model and the protein structure of Baoshan pig IGF-1 gene was-1.15, which has good consistency. The study indicates that IGF-1 gene is highly conserved in the process of species evolution,so that its amino acid sequence encoding Baoshan pig and Duroc pig is the same,and there is little or no difference in protein structure and function.Whether IGF-1 gene can be used as a candidate gene to regulate the lean meat,fat and slow growth of Baoshan pig still needs to be studied.
