银杏内生真菌CS15菌株鉴定与杀螺作用的初步研究

摘  要:目的:研究可能影响银杏内生真菌CS15菌株杀螺效果的相关因素。方法:应用形态学特征观察结合rDNA-ITS序列综合分析对该菌株进行鉴定。使用浸泡法配制不同浓度的菌株CS15发酵液;实验随机分为4组(超纯水对照组和1%、2%、4%滤液组),评定不同浓度CS15发酵滤液对钉螺的杀灭效果。采用蒽酮比色定量分析法检测发酵滤液浸泡处理后的螺体内糖原含量变化。结果:菌株经分离培养、形态学及分子生物学鉴定,该菌株为草酸青霉真菌。实验组菌株CS151%、2%、4%浓度下的发酵滤液均有一定的杀螺效果。其中在24 h后,以4%发酵滤液杀螺效果最佳(P<0.05),在48 h和72 h后,4%发酵滤液杀螺效率优于1%和2%发酵滤液(P<0.05)。经不同浓度CS15发酵滤液处理后,钉螺软体组织内糖原含量均明显降低,以4%发酵滤液组最为明显(P<0.05)。结论:在本实验条件下,银杏内生真菌CS15发酵滤液对湖北钉螺具有毒杀效应,以4%发酵滤液杀螺效果最佳,其杀螺因素可能与螺软体组织内糖原合成相关。Objective:To investigate the related factors that potentially affect the molluscidical efficacy of endophytic fungus CS15 strain isolated from Ginkgo biloba.Methods:The strain was identified via morphological observation and rDNA-ITS sequence analysis.Immersion method was used to prepare the fermentation filtrate of strain CS15 with different concentrations.The experiment was randomly carried out in 4 groups(ultra-pure water control group and 1%,2%and 4%filtrate group)to evaluate the killing effect of different concentration of CS15 filtrate on O.hupensis.Anthrone colorimetric method was used to measure the change of glycogen content in snail after soaking in fermentation filtrate.Results:The isolated strain was identified as Penicillium oxalicum via culturing,morphological observation and molecular biology analysis.The fermentation filtrate of strain CS15 at 1%,2%and 4%concentration had certain molluscicidal activities.The efficacy of snail killing was the best after 24 h by 4%fermentation filtrate(P<0.05),and the killing efficiency of 4%fermentation filtrate was better than that 1%and 2%fermentation filtrate after 48 h and 72 h(P<0.05).After treatment with different concentration of CS15 fermentation filtrate,the glycogen content in mollusca tissues of O.hupensis was significantly decreased,especially in 4%fermentation filtrate group(P<0.05).Conclusion:Under current experimental conditions,the fermentation filtrate of endophytic fungus CS15 isolated from Ginkgo biloba may produce toxic effect on O.hupensis,and the killing efficacy is optimal at 4%of the fermentation filtrate.The potential mechanism of snail killing might be associated with glycogen synthesis of CS15 in the soft tissues of snail.
