摘  要:【目的】明确不同苗种来源香港牡蛎的生长差异及生长相关基因表达模式,为筛选出生长性状候选基因打下理论基础,也为繁育优质香港牡蛎新品种提供参考依据。【方法】通过比较人工繁育(人工组)和自然海域半人工采苗(天然组)2种模式获得香港牡蛎的生长差异,利用实时荧光定量PCR检测不同苗种来源香港牡蛎外套膜和闭壳肌中IGF1、AMPKα和FoxO1基因的表达情况,并利用统计学方法及相关分析探究生长指标(壳高、壳长、壳宽、鲜重和软体重)与生长相关基因表达变化间的相关性。【结果】人工组和天然组香港牡蛎在试验期间(4月龄~12月龄)的壳高、壳长、壳宽、鲜重和软体重等生长指标整体上呈增长趋势;人工组香港牡蛎的生长指标均显著高于天然组香港牡蛎(P<0.05,下同),但天然组香港牡蛎生长指标的整体增幅高于人工组香港牡蛎。天然组香港牡蛎外套膜和闭壳肌中IGF1、AMPKα和FoxO1基因在不同发育阶段的相对表达量整体上高于人工组香港牡蛎,但差异均不显著(P>0.05,下同)。相关分析结果表明,人工组和天然组香港牡蛎生长相关基因表达与生长性状间具有相关性,其中外套膜和闭壳肌的IGF1基因表达总量与壳高呈负相关,与壳宽呈正相关;AMPKα基因表达总量与壳高、鲜重和软体重等生长指标呈正相关,与壳宽呈负相关;FoxO1基因表达总量与壳宽呈负相关。【结论】无论是人工繁育还是自然海域半人工采苗获得的香港牡蛎,在生长性状及相关基因表达方面均存在一定差异。其中,IGF1、AMPKα和FoxO1基因参与调控香港牡蛎的生长发育,且对不同生长指标有不同的指示作用,因此开展人工繁育时可通过调控这些基因的表达以改变香港牡蛎生长指标的增幅。【Objective】Growth differences during the different seedling sources of Crassostrea hongkongensis and their correlations with the expression patterns of growth genes were investigated to provide a theoretical basis for screening candidate genes for growth traits and provide reference for breed selection of C.hongkongensis.【Method】Growth dif-ferences were observed by comparing C.hongkongensis derived from artificial spat-collection(artificial group)and semi artificial collection of seedling in natural ocean systems(natural group).The expression of IGF1,AMPKαand FoxO1 genes in the mantles and adductor muscles of C.hongkongensis was deduced by real-time fluorescence quantitative PCR.Correlation was analyzed between growth traits(shell height,shell length,shell width,whole weight and soft tissue weight)and growth-related gene expression by statistical methods and correlation analysis.【Result】Growth traits including shell height,shell length,shell width,whole weight and soft tissue weight of C.hongkongensis obtained from artificial group and natural group showed an overall increasing trend during the experiment(4-month-old to 12-month-old).The growth traits of artificial group were significantly higher than natural group(P<0.05,the same below).However,the growth rate of natural group was higher than that of artificial group.The gene expression of IGF1,AMPKαand FoxO1 in the mantle and adductor muscle in natural group were higher than those in artificial group at different developmental stages,but the differences were not significant(P>0.05,the same below).Correlation analysis results showed that there was correlation between growth-related genes and growth traits of the artificial group and natural group.The total expres-sion of IGF1 in the mantle and adductor muscle was negatively correlated with shell height and positively correlated with shell width.The total expression of AMPKαwas positively correlated with growth traits including shell height,whole weight and soft tissue weight,and negatively correla
