摘  要:目的研究绞股蓝总皂苷和绞股蓝不同提取物对大鼠血压的影响。方法将大鼠分为9组,除空白对照组外,高血压模型组、绞股蓝总皂苷对照组、绞股蓝总皂苷干预组(绞股蓝总皂苷180 mg/kg、360 mg/kg、720 mg/kg,低、中、高剂量组)、总提取物组、乙酸乙酯提取物组和正丁醇提取物组的大鼠均被喂养高脂高糖饲料和饮用糖盐水(糖4%、盐1%)和果糖水(6%)。绞股蓝总皂苷对照组、绞股蓝总皂苷干预组、总提取物组、乙酸乙酯提取物组和正丁醇提取物组的大鼠均给予相应剂量的药物灌胃。给药8周后处死大鼠,测定大鼠血清中一氧化氮(NO)、内皮素(ET)和血管紧张素Ⅱ(AngⅡ)含量。结果与空白对照组相比,高糖高脂饮食能明显使大鼠血压的增高[收缩压增高(34.9±9.8)%,舒张压增高(17.9±8.5)%],差异有统计学意义(P<0.05)。与高血压模型组相比,绞股蓝总皂苷对照组收缩压降低(39.7±6.9)%、舒张压降低(36.7±7.3)%,血压显著性降低(P<0.05)。与空白对照组相比,绞股蓝总皂苷对照组、乙酸乙酯提取物组、正丁醇提取物组收缩压、舒张压相差不大(P>0.05)。绞股蓝总皂苷明显降低大鼠的血压,同时能显著增加大鼠血清NO含量,降低大鼠血清ET和AngⅡ的含量。结论绞股蓝总皂苷及其乙酸乙酯、正丁醇提取物可预防高糖高脂诱导的大鼠血压的增高。绞股蓝总皂苷可能通过降低ET、AngⅡ含量和升高NO含量来发挥对大鼠的降压作用。Objective To investigate the anti-hypertensive effects of different extracts from Gynostemma pentaphylla in experimental hypertensive rats.Methods The rats were divided into 9 groups,except the rats in blank control group,those in the hypertensive model group,Gypenosides control group,Gypenosides intervention group,total extract group,ethyl acetate extract group,and n-butanol extract group were fed with high-fat and high sugar diet and sugar saline(sugar 4%,salt 1%)and fructose water(6%).Rats in the Gypenosides control group,Gypenosides intervention group,total extract group,ethyl acetate extract group,and n-butanol extract group were given with corresponding doses of drugs by gavage.After 8 weeks of administration,the rats were sacrificed and the contents of NO,ET and AngⅡin serum were measured.Results Compared with the blank control group,the high sugar and high fat diet could significantly increase the blood pressure of rats[systolic blood pressure increased by(34.9±9.8)%,diastolic blood pressure increased by(17.9±8.5)%]with significant difference(P<0.05).Compared with the hypertension model group,the systolic blood pressure and diastolic blood pressure of the Gypenosides control group significantly decreased by(39.7±6.9)%and(36.7±7.3)%,respectively(P<0.05).Compared with the blank control group,there was no significant difference in the systolic and diastolic pressure in Gypenosides control group,ethyl acetate extract group or n-butanol extract group(P>0.05).The total saponins of Gynostemma pentaphyllum can significantly reduce the blood pressure of rats,increased the content of NO in serum of rats,and reduced the content of ET and AngⅡin serum of rats.Conclusion Ethyl acetate group and N-butanol group of Gynostemma pentaphyllum can prevent the increase of blood pressure induced by high glucose and high fat in rats,and also have a relatively safe curative effect on hypertension.Gypenosides may exert antihypertensive effect on rats by reducing the contents of ET and AngⅡand increasing the content of N
