红核妇洁洗液酮类成分治疗外阴阴道假丝酵母菌病的作用和机制研究

摘　　要：目的探讨红核妇洁洗液酮类成分治疗外阴阴道假丝酵母菌病(vulvovaginal candidiasis,VVC)的作用和机制。方法2021年12月至2022年6月于西安交通大学医学部通过网络药理学和生物信息学相关手段,筛选红核妇洁洗液酮类成分目标靶点和VVC疾病靶点,分析交集靶点;应用VVC大鼠模型观察红核妇洁洗液对大鼠阴道黏膜的作用,检测关键靶点的蛋白表达与正常组比较,模型组大鼠阴道组织中STAT1蛋白表达显著升高50%左右(P<0.001);与模型组相比,红核妇洁洗液组及氟康唑组大鼠阴道组织STAT1蛋白表达显著降低约48%(P<0.001)。。结果红核妇洁洗液中酮类成分共30个,酮类成分治疗VVC共有98个靶点,牡荆素、山奈酚具有最多的潜在靶点,STAT1为靶点网络中的关键靶点。苏木精-伊红(HE)染色结果显示,红核妇洁洗液可改善大鼠阴道黏膜损伤;Western blotting结果显示,红核妇洁洗液降低了白假丝酵母菌诱导的阴道组织中STAT1的表达。进一步富集发现,酮类成分主要可调控JAK-STAT1通路相关靶标。结论红核妇洁洗液中酮类主要活性成分可能为牡荆素、山奈酚,红核妇洁洗液可通过调控STAT1蛋白表达发挥抗炎抑菌的作用,治疗VVC。Objective To investigate the effect and mechanism of ketone components in Honghe Fujie lotion in the treatment of vulvovaginal candidiasis.Methods The targets of ketones in Honghe Fujie lotion for the treatment of VVC were screened by related means of network pharmacology and bioinformatics and the intersection targets were analyzed.The VVC rat model was used to observe the effect of Honghe Fujie lotion on vaginal mucosa of rats and the protein expression of key targets was detected in the Xi’an Jiaotong University Health Science Center from December 2021 to June 2022.Results There were 30 ketones in Honghe Fujie lotion.There were 98 targets for the treatment of VVC by ketones.Vitexin and kaempferol had the most potential targets.STAT1 was the key target in the target network.HE staining results showed that Honghe Fujie lotion could improve vaginal mucosa injury in rats.Western blotting results showed that Honghe Fujie lotion reduced the expression of ST AT1 in vaginal tissue induced by Candida albicans.Further enrichment found that ketones mainly regulated JAK-STAT1 pathway-related targets.Conclusion The main active ingredients in the ketones of Honghe Fujie lotion may be vitexin and kaempferol,which can play an anti-inflammatory and antibacterial role by regulating the expression of STAT1 protein and treat vulvovaginal candidiasis.
