从线粒体质量控制探讨中医药防治神经系统疾病的新靶点

摘　　要：中医药对神经系统疾病的治疗作用与线粒体质量控制机制密切相关。中医药对线粒体干预的药理机制主要集中于线粒体呼吸功能、线粒体膜电位、线粒体氧化损伤、线粒体钙调节功能、线粒体凋亡途径等方面,对线粒体质量控制的研究多集中在对线粒体生物合成的影响方面,认为线粒体生物合成可能是中医药防治神经系统疾病的关键,缺少对线粒体动力学、线粒体自噬及完整控制链的研究。此外,目前的研究还存在一些问题,如中医药调控线粒体自噬治疗脑血管疾病的相关研究质量偏低,大多只涉及线粒体自噬通路相关蛋白的表达水平,并未证实线粒体自噬是否是中医药治疗脑血管疾病的关键机制。今后可深入探讨线粒体质量控制在神经系统疾病的病理生理进程中的作用机制,全面阐释中医药保护线粒体的作用机制,为临床治疗及新药开发提供科学依据。The therapeutic effect of TCM on nervous system diseases is closely related to the mechanism of mitochondrial quality control.The pharmacological mechanism of TCM intervention on mitochondria mainly focuses on mitochondrial respiratory function,mitochondrial membrane potential,mitochondrial oxidative damage,mitochondrial calcium regulation function,mitochondrial apoptosis pathway,etc.And its research on mitochondrial quality control is still in its infancy,mostly focusing on the impact on mitochondrial biosynthesis,and lacking research on mitochondrial dynamics,mitophagy,and the complete control chain.In addition,there are still some problems in the carrent research.For example,the research on the regulation of mitophagy by traditional Chinese medicine in the treatment of cerebrovascular diseases is not deep enough and the quality is low.Most of them only involve the expression level of proteins related to the mitophagy pathway.The TCM intervention mechanism on mitochondrial quality control has great research value.In the future,the mechanism of mitochondrial quality control in the pathophysiological process of nervous system diseases can be further explored,thus comprehensively explaining the TCM protection mechanism of mitochondria,and providing a scientific basis for clinical treatment and new drug development.
