摘 要:目的:观察清肺络咳饮对咳嗽变异性哮喘豚鼠白细胞介素-5(interleukin-5,IL-5)、IL-13、转化生长因子-β1(transforming growth factor-β,TGF-β1)的影响。方法:将90只SPF级雄性豚鼠随机分为6组,即空白组、模型组、地塞米松组及清肺络咳饮高(4.8 g·kg^(-1))、中(2.4 g·kg^(-1))、低(1.2 g·kg^(-1))剂量组,每组15只。除空白组外,其余各组豚鼠采用OVA致敏和激发,并以Al(OH)3为免疫佐剂的方法建立动物模型。造模的同时,豚鼠灌胃给予相应药物,空白组和模型组给予等体积生理盐水灌胃,每日1次,持续14 d。药物干预结束后,观察各组豚鼠咳嗽次数;瑞氏吉姆萨染色观察肺泡灌洗液(bronchoalveolar lavage fluid,BALF)中的细胞分类并计数;ELISA法检测BALF中白细胞介素-5(interleukin-5,IL-5)、IL-13和转化生长因子-β1(transforming growth factor-β1,TGF-β1)的水平;Western Blot检测肺组织中IL-5、IL-13和TGF-β1蛋白表达水平。结果:与空白组比较,模型组豚鼠咳嗽次数显著增多(P<0.05),BALF中细胞总数、嗜酸性粒细胞数、淋巴细胞数、巨噬细胞数、中性粒细胞数显著增多(P<0.05),BALF及肺组织中IL-5、IL-13、TGF-β1的水平明显升高(P<0.05);与模型组比较,清肺络咳饮各剂量组豚鼠咳嗽次数明显减少(P<0.05),BALF中细胞总数、嗜酸性粒细胞数、巨噬细胞数、中性粒细胞数、淋巴细胞数显著减少(P<0.05),BALF及肺组织中IL-5、IL-13、TGF-β1的水平明显降低(P<0.05)。结论:清肺络咳饮可改善咳嗽变异性哮喘豚鼠的气道高反应性和气道炎症反应,且该作用可能与调控IL-5、IL-13、TGF-β1的表达有关。Objective:To observe the effect of Qingfei Luoke Decoction on interleukin-5(IL-5),IL-13,transforming growth factor-β1(TGF-β1)Impact in guinea pigs with cough variant asthma(CVA).Methods:90 SPF male guinea pigs were randomly divided into 6 groups:blank group,model group,dexamethasone group,and Qingfei Luoke Decoction high(4.8 g·kg^(-1)),medium(2.4 g·kg^(-1)),low(1.2 g·kg^(-1))dose group,15 rats in each group.Except for the blank group,the other groups of guinea pigs were sensitized and stimulated with OVA,and the animal model was established by using Al(OH)3 as an immune adjuvant.Simultaneously with modeling,guinea pigs were intragastrically administered corresponding drugs,and the blank group and model group were intragastrically administered equal volumes of normal saline,once a day for 14 days.After the drug intervention,the frequency of coughing in guinea pigs in each group was observed.Wright′s Giemsa staining was used to observe the cell classification and count in the bronchoalveolar lavage fluid(BALF).The levels of interleukin-5(IL-5),IL-13,and transforming growth factor-β1(TGF-β1)in BALF were detected by the ELISA method.And the protein expression levels of IL-5,IL-13,and TGF-β1 in lung tissue were detected by Western Blot.Results:Compared with the blank group,the number of coughs in guinea pigs significantly increased(P<0.05).The total number of cells,eosinophils,lymphocytes,macrophages,and neutrophils in the BALF were significantly increased(P<0.05),and the levels of IL-5,IL-13,and TGF-β1 in BALF and lung tissue were significantly increased(P<0.05).Compared with the model group,the number of coughs in guinea pigs in each dose group of Qingfei Luoke Decoction was significantly reduced(P<0.05),and the total number of cells,eosinophils,macrophages,neutrophils,and lymphocytes in BALF were significantly decreased(P<0.05),and the levels of IL-5,IL-13,and TGF-β1 in BALF and lung tissue were significantly reduced(P<0.05).Conclusion:Qingfei Luoke Decoction can improve airway hyperresponsiveness and a
