摘 要:目的研究黄香喷雾剂对紫外线损伤大鼠皮肤的保护作用。方法将SD大鼠随机分为7组:空白对照组、紫外线模型组、基质组、阳性药物组和黄香喷雾剂低、中、高剂量组,除空白对照组外,其余各组大鼠进行UV(UVA联合UVB)照射建立紫外线损伤模型,实验周期为9周,总照射强度:UVB为14.13 J/cm^(2),UVA为61.23 J/cm^(2)。观察大鼠背部皮肤受损情况并进行评分;采用HE、Masson染色并用透射电镜观察大鼠皮肤组织病理变化;采用酶联免疫吸附法(ELISA)测定大鼠皮肤组织中的超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、过氧化氢酶(CAT)和透明质酸(HA)含量。结果与空白对照组相比,紫外线模型组大鼠背部皮肤受损严重,有明显起皮和干裂现象,皮肤角质层角化过度伴角化不全,表皮层增厚,真皮层坏死,炎性细胞浸润,胶原纤维排列不规则,且真皮层成纤维细胞线粒体发生肿胀,粗面内质网见少量自噬现象,说明建模成功,经紫外线照射后大鼠皮肤组织中的SOD、GSH-Px、CAT及HA含量显著降低(P<0.05);与紫外线模型组相比,黄香喷雾剂低、中、高剂量组大鼠皮肤损伤程度有所减轻,皮肤角化明显,表皮层结构相对完整,真皮层被少量炎性细胞浸润,胶原纤维排列较整齐,成纤维细胞内的线粒体肿胀程度相对减轻,皮肤组织中的SOD、GSH-Px、CAT、HA含量显著升高(P<0.05)。结论黄香喷雾剂对紫外线照射所致大鼠皮肤损伤有一定的保护作用,其作用机制可能与抗氧化有关。Objective To study the protective effect of Huangxiang spray on skin of rats damaged by ultraviolet radiation.Methods SD rats were randomly divided into 7 groups:blank control group,UV model group,substrate group,positive drug group(VE),low-dose,medium-dose and high dose groups of Huangxiang spray,except for the blank control group,the other groups were irradiated with UV(UVA combined with UVB)to establish ultraviolet damage model,the experimental period was 9 weeks,and the total irradiation intensity:UVB was 14.13 J/cm^(2),UVA was 61.23 J/cm^(2).the skin damage on the back of rats was observed and scored;the pathological changes of skin tissue were observed by HE and Masson staining and transmission electron microscopy;the contents of superoxide dismutase(SOD),glutathione peroxidase(GSH-Px),catalase(CAT)and hyaluronic acid(HA)in rat skin were determined by enzyme-linked immunosorbent assay(ELISA).Results Compared with the blank control group,the back skin of rats in the UV model group was seriously damaged,with obvious peeling and dry cracking,hyperkeratosis of the stratum corneum with incomplete keratosis,thickening of the epidermis,necrosis of the dermis,infiltration of inflammatory cells,irregular arrangement of collagen fibers,and a small amount of autophagy in the rough endoplasmic reticulum,indicating that the modeling was successful,after UV irradiation,the contents of SOD,GSH-Px,CAT and HA in skin tissue of rats were significantly reduced(P<0.05);compared with the UV model group,the degree of UV damage in the skin of rats in the low,medium and high dose groups of Huangxiang spray was alleviated,skin keratinization was obvious,the structure of the epidermis was relatively intact,the dermis was infiltrated by a small number of inflammatory cells,the collagen fibers were arranged neatly,the swelling of mitochondria in fibroblasts was relatively reduced,the contents of SOD,GSH-Px,CAT and HA in skin tissue were significantly increased(P<0.05).Conclusion Huangxiang spray has a certain protective effect on skin
