摘 要:目的分析青海地区藏族、汉族毒性弥漫性甲状腺肿(Graves disease,GD)患者与健康者血清巨噬细胞移动抑制因子基因(Macrophage migration inhibitory factor,MIF)rs755622、rs1007888、rs2096525位点基因多态性,探讨其与青海藏族、汉族GD发病的关联性。方法本研究设GD组(汉族60例,藏族60例)、对照组(汉族60例,藏族60例),应用SNaPshot法测定MIF基因多态性。结果藏族、汉族GD组及对照组均符合Hardy-Weinberg平衡检验,提示样本具有良好群体代表性。藏族GD组、藏族对照组、汉族GD组、汉族对照组四组MIF-rs755622位点基因型(GG、GC、CC)、rs1007888位点基因型(GG、GA、AA)、rs2096525位点基因型(TT、TC、CC)频率的差异均无统计学意义;各位点等位基因(G/C、G/A、T/C)差异均无统计学意义。结论MIF-rs755622位点GC单核苷酸多态性、rs1007888位点GA单核苷酸多态性、rs2096525位点TC单核苷酸多态性均与青海地区藏族、汉族GD发病无关联,该基因不是青海地区藏族、汉族GD发病的易感候选基因。Objective To analyze the gene polymorphisms of rs755622,rs1007888 and rs2096525 of the serum macrophage migration inhibitory factor gene in Tibetan and Han patients with Graves disease and healthy individuals in Qinghai province,and to explore their relationship between the incidence of GD in Tibetan and the Han nationalities.Methods A total of 120 cases in the GD group(60 Han and 60 Tibetan)and 120 cases in the control group(60 Han and 60 Tibetan)were enrolled in this study.The SNaPshot method was used to determine the MIF gene polymorphisms.Results The GD group and the control group of Tibetan and Han nationalities all conform to the Hardy Weinberg balance test,suggesting that the sample has good population representation.There was no significant diference in the frequencies of MIF-rs755622 locus genotype(CG,GC,CC),rs1007888 locus genotype(GG,GA,AA),rs2096525 locus genotype(TT,TC,CC)among Tibetan GD group,Tibetan control group,Han GD group and Han control group;There was no significant difference in alleles(G/C,G/A,T/C)at each locus.Conclusion MIF-rs755622 GC single nucleotide polymorphism,rs1007888 GA single nucleotide polymorphism,rs2096525 CT single nucleotide polymorphism are not associated with the incidence of GD in Tibetan and Han ethnic groups in Qinghai.This gene is not a susceptibility candidate gene for GD in Tibetan and Han nationalities in Qinghai.
