摘 要:目的探讨山萘酚对兔耳增生性瘢痕(HS)的抑制作用。方法4月龄新西兰大耳白兔8只,4只雄兔,4只雌兔,体质量约(2.5±0.2)kg,体质量范围为2.3~2.7 kg。采用随机数表法将8只大耳白兔随机分为山萘酚组和氯化钠组,每组4只。兔耳瘢痕模型建模成功后,对组织进行大体观察,并行HE染色、Masson染色、免疫组化检测、Western-blot检测测定兔耳瘢痕组织内蛋白激酶B(Akt)和磷酸化蛋白激酶B(p-Akt)的表达水平。结果给药7 d后,山萘酚组的瘢痕较氯化钠组的瘢痕颜色更淡,表面更平,触感稍软。HE染色可见山萘酚组表皮层及真皮层较氯化钠组稍薄、无明显增厚,炎症细胞可见明显减少,胶原纤维排列较整齐规律且有序,成纤维细胞呈纺锤形、细长且排列较为规整。Masson染色可见氯化钠组过度堆积的胶原纤维,山萘酚组可见胶原纤维束且胶原厚度较氯化钠组更薄、排列更整齐。免疫组化检测可见山萘酚组兔耳瘢痕p-Akt表达明显低于氯化钠组。Western-blot检测可见p-Akt表达明显低于氯化钠组,但Akt的表达未见明显差异。结论山萘酚可通过抑制p-Akt的表达水平来抑制兔耳瘢痕形成。Objective To investigate the therapeutic effect of kaempferol on hypertrophic scar of rabbit ear.Methods A total of 84-months-old New Zealand white rabbits(4 male rabbits,4 female rabbits)with body weight with body weight of(2.5±0.2)kg,ranging from 2.3 to 2.7 kg were obtained.A total of 8 large eared white rabbits were randomly divided into experimental group and control group by random number table method,with 4 rabbits in each group.After the scar model of rabbit ear was built successfully,the tissue was observed in general,and the expression levels of protein kinase B(Akt)and phosphorylated protein kinase B(p-Akt)in rabbit ear scar tissue were determined by HE staining,Masson staining,immunohistochemical detection and Western-blot detection.Results After 7 days of administration,scars in the experimental group were lighter in color,flatter in surface,and slightly softer to the touch than scars in the control group.HE staining showed that the epidermis and dermis of the experimental group were slightly thinner than that of the control group,without obvious thickening,inflammatory cells were significantly reduced,collagen fibers were arranged in a regular and orderly manner,and fibroblasts were fusiform,slender and orderly.Masson staining showed excessive accumulation of collagen fibers in the control group,and collagen fiber bundles in the experimental group were thinner and more neatly arranged than in the control group.Immunohistochemical test showed that the expression of p-Akt in rabbit ear scar in the experimental group was significantly lower than that in the control group.Western-blot analysis showed that the expression of p-Akt was significantly lower than that of the control group,but there was no significant difference in the expression of Akt.Conclusions Kaempferol can inhibit ear scarring by inhibiting the expression level of p-Akt.
