摘 要:目的:确定川贝母主要活性部位总生物碱的纯化条件,为其开发利用和产业化生产提供理论依据。方法:以解析液中的总生物碱含量为指标,分别考察H103型大孔吸附树脂和001x7型离子交换树脂对川贝母提取物的最佳吸附及洗脱条件。结果:经H103型大孔吸附树脂纯化后,所得产物的纯度和收率分别为44.3%和0.71%。而经001x7型离子交换树脂纯化后,所得产物的纯度和收率分别为61.4%和1.24%。因此001x7型离子交换树脂对川贝母总生物碱的纯化效果最好,其最佳纯化条件:上样液pH值为2,上样体积为0.75 BV,水洗体积为8 BV,用含5%氯化钠的60%乙醇洗脱,洗脱剂用量为10BV。结论:两种纯化工艺都具有简便、快速、准确等优点,都可用于川贝母总生物碱的富集纯化。但001x7型离子交换树脂对川贝母总生物碱纯化效果更好。Objective:The purification conditions of main active complementary total alkaloids from Fritillaria Cirrhosae Bulbs were determined to provide theoretical basis for its development and industrial production. Methods:The optimal adsorption and elution conditions for Fritillaria Cirrhosae Bulbs extracts were investigated by H103 macroporous adsorption resin and 001x7 ion exchange resin, respectively, using the total alkaloid content in the analytical solution as index. Results:After purification by H103 macroporous resin, the purity and yield of the product were 44.3% and 0.71%,respectively. After purification by 001x7 ion exchange resin, the purity and yield of the product were 61.4% and 1.24%,respectively. Therefore, 001x7 ion exchange resin has the best purification effect on the total alkaloid of Fritillaria Cirrhosae Bulbs. The optimal purification conditions are as follows: pH value of sample solution is 2,sample volume is 0.75 BV,water volume is 8BV,eluted with 60% ethanol containing 5% sodium chloride, eluent dosage is 10BV. Conclusion:Both purification processes are simple, rapid and accurate, and can be used for the enrichment and purification of total alkaloids from Fritillaria Cirrhosae Bulbs. But 001x7 ion exchange resin has better purification effect on the total alkaloids of Fritillaria Cirrhosae Bulbs.
