摘  要:人参根腐病是一种土传性真菌病害,是人参根部主要病害之一,严重影响着人参产业的可持续健康发展。为筛选与人参抗根腐病性状相关联的SSR标记,本研究以人参抗根腐病株系和感根腐病株系的基因组DNA为模板,采用琼脂糖凝胶电泳和聚丙烯酰胺凝胶电泳两种方法,对实验室前期所筛选出的18对SSR多态性引物进行了人参根腐病抗病性分子标记的筛选,结果表明,引物P29可以将人参抗根腐病和感根腐病株系区分开来,该标记可能与人参根腐病抗性相关,可作为鉴定人参根腐病抗病性的分子标记,用于人参根腐病早期筛选。该研究结果为后续开展人参根腐病基因定位及抗病育种提供了良好的基础,为人参抗病性状的分子检测及分子标记辅助选择育种提供有益参考,推动了分子标记在人参育种中的应用。Ginseng root rot is a soil-borne fungal disease,which is one of the main root diseases of ginseng.It seriously affects the sustainable and healthy development of ginseng industry.In order to screen SSR markers associated with root rot resistance traits of ginseng,genomic DNA of ginseng root rot resistant and susceptible strains was used as a template in this study.Two methods of agarose gel electrophoresis and polyacrylamide gel electrophoresis were used.The 18 pairs of SSR polymorphic primers screened in the previous laboratory were used to screen molecular markers for the resistance of ginseng root rot disease.The results showed that primer P29 could distinguish root rot resistance and susceptible strains of ginseng,and this marker might be related to root rot resistance of ginseng.It could be used as a molecular marker to identify the resistance of ginseng root rot and for early screening of ginseng root rot.The results of this study laid a good foundation for the future development of ginseng root rot gene mapping and disease resistance breeding.It provides a useful reference for molecular detection of disease resistance traits and marker-assisted selection breeding of ginseng.And it will promote the application of molecular markers in ginseng breeding.
