毛莨科植物总状升麻活性成分对运动大鼠能量代谢的影响

摘  要:毛茛科药用植物总状升麻(Cimicifuga racemosa L.)是一种传统中药,其提取物中的主要活性成分为三萜皂苷(23-epi-26-Deoxyactein)。为了揭示总状升麻提取物活性成分23-epi-26-Deoxyactein (DA)对大鼠的运动能力和能量代谢的影响,本研究连续4周对力竭游泳运动大鼠灌胃5、10和20 mg·kg^(-1)·d^(-1)的23-epi-26-Deoxyactein,研究结果显示,23-epi-26-Deoxyactein剂量依赖性地升高了大鼠的力竭游泳时间(P<0.05),降低了血清CK、ALT和AST水平(P<0.05),升高了肝糖原和肌糖原含量(P<0.05),降低了血清LA和BUN水平,升高血清NEFA水平(P<0.05)。23-epi-26-Deoxyactein剂量依赖性地升高了骨骼肌SOD和CAT水平(P<0.05),降低了骨骼肌MDA水平(P<0.05)。23-epi-26-Deoxyactein剂量依赖性地升高了骨骼肌PGC-1α、TFAM、NRF1和ATP5B的mRNA相对表达量和p-AMPKα的蛋白相对表达量(P<0.05)。本研究初步表明总状升麻活性成分23-epi-26-Deoxyactein提高了力竭运动大鼠的运动能力并改善了能量代谢,其机制可能与AMPK信号通路的激活有关。Cimicifuga racemosa L.,a medicinal plant of the Ranunculaceae family,is a traditional Chinese medicine.The main active ingredient in the extract is triterpenoid saponin(23-epi-26-Deoxyactin).To explore the effects of C.racemosa active component 23-epi-26-Deoxyactein(DA)on exercise capacity and energy metabolism in rats,this study gavaged 23-epi-26-Deoxyactin at 5,10 and 20 mg·kg^(-1)·d^(-1) for 4 weeks in rats after exhaustive swimming.The results showed that 23-epi-26-Deoxyactin dose-dependently increased the exhaustive swimming time of rats(P<0.05),and decreased the serum CK,ALT and AST levels(P<0.05),it increased the content of liver glycogen and muscle glycogen(P<0.05),decreased the level of serum LA and BUN,and increased the level of serum NEFA(P<0.05).23-epi-26-Deoxyactin dose-dependently increased the level of SOD and CAT in skeletal muscle(P<0.05),and decreased the level of MDA in skeletal muscle(P<0.05).23-epi-26-Deoxyactin dosedependently increased skeletal muscle PGC-1α、Relative mRNA expression and p-AMPKαof TFAM,NRF1 and ATP5B,the relative expression level of the protein was(P<0.05).This study preliminarily showed that 23-epi-26-Deoxyactin,the active component of C.racemosa,improved the exercise ability and energy metabolism of rats after exhaustive exercise,and its mechanismmay be related to the activation ofAMPKsignal pathway.
