摘  要:以牛樟芝为试验材料,采用单因素和正交试验的设计方案,研究不同碳源(乳糖、蔗糖、葡萄糖、可溶性淀粉、甘露醇)和氮源(黄豆粉、酵母粉、牛肉膏、蛋白胨、麦芽浸膏)对菌丝体生长的影响。通过观测其菌落大小、菌落长势、菌丝生长速度及生长指数,筛选出最适宜牛樟芝菌丝体生长的培养条件。结果显示,在单因素试验中葡萄糖是牛樟芝菌种的最适碳源,酵母粉是最适氮源,该条件下其菌丝密度高,颜色呈橘红色,生长速度快。以菌丝生长速度和生长指数作为评价指标的正交试验中,当培养基配方为马铃薯(煮汁) 200 g·L^(-1)、葡萄糖20 g·L^(-1)、酵母粉15 g·L^(-1)、KH_(2)PO_(4)1 g·L^(-1)、MgSO_(4)0.5 g·L^(-1)、维生素B10.1 g·L^(-1)时,牛樟芝菌丝生长速度达2.47 mm·d^(-1),生长指数为12.35,明显优于其他试验组。Taking Antrodia camphporata as the material,and the single factor and orthogonal experiment as the method,to study the effect of different carbon source(lactose,sucrose,glucose,soluble starch,mannitol)and nitrogen source(soybean flour,yeast powder,beef extract,peptone,malt extract)on mycelium growth.By observing the colony size,colony development,mycelial growth rate and growth index,the optimum culture conditions for mycelial growth of A.camphporata were explored.The results showed that glucose is the most suitable carbon source and yeast powder is the most suitable nitrogen source in single factor experiment,and the mycelium had high density,orange-red in color and fast growth rate.In orthogonal experiment with mycelial growth rate and growth index as evaluation indexes.When the medium froumla is potato(boiled to liquor)200 g·L^(-1),glucose 20 g·L^(-1),yeast powder 15 g·L^(-1),KH_(2)PO_(4)1 g·L^(-1),MgSO_(4)0.5 g·L^(-1),vitamin B10.1 g·L^(-1),the mycelial growth rate of A.camphporata reached 2.47 mm·d^(-1) and the growth index was 12.35,which was obviously better than other groups.
