内皮细胞特异性分子1与冠心病大鼠主动脉内皮细胞炎性损伤和血管重构的关系

摘　　要：目的探究内皮细胞特异性分子1(Endocan)与冠心病大鼠主动脉内皮细胞炎性损伤和血管重构的关系及Nod样受体蛋白3(NLRP3)信号通路的作用机制。方法选择雄性健康SD大鼠40只,随机分成模型组和正常组(n=20),对比2组血清Endocan、白细胞介素(IL)10、IL-1β,主动脉内皮组织Endocan、NLRP3表达。将主动脉内皮细胞分成正常对照组(正常大鼠细胞培养获得)、阳性对照组(模型组大鼠内皮细胞)、Endocan病毒组(转染Endocan慢病毒)和Endocan-shRNA病毒组(Endocan沉默干扰病毒),采用小管形成实验检测各组细胞成管能力。结果模型组大鼠血钙、TC、TG、LDL、Endocan、IL-10、IL-1β及主动脉内皮组织Endocan、NLRP3 mRNA和蛋白表达明显高于正常组,差异有统计学意义(P<0.01)。Endocan病毒组内皮细胞管腔形成数目、IL-6、IL-10、IL-1β和IL-18水平及内皮细胞NLRP3 mRNA和蛋白表达明显高于正常对照组、阳性对照组、Endocan-shRNA病毒组,差异有统计学意义(P<0.05)。Endocan-shRNA病毒组内皮细胞管腔形成数目、IL-6、IL-10、IL-1β和IL-18水平明显高于正常对照组,差异有统计学意义(P<0.05)。Endocan-shRNA病毒组内皮细胞NLRP3 mRNA和蛋白表达明显高于正常对照组和阳性对照组,差异有统计学意义(5.55±1.19 vs 4.56±1.19,4.98±1.03;1.34±0.27 vs 0.78±0.12,1.09±0.21,P<0.05)。结论Endocan的过表达可促进主动脉内皮细胞炎性损伤,激活血管重构,并与NLRP3信号通路的表达有关。Objective To explore the relationship of Endocan with inflammatory injury and vascular remodeling of aortic endothelial cells in rats with coronary heart disease and the mechanism of NLRP3 inflammasome signaling pathway.Methods A total of 40 male healthy SD rats were randomly divided into model group and normal group(n=20).The abdominal aortic blood was collected to compare the serum levels of Endocan,IL-10,IL-1,and the mRNA and protein levels of Endocan and NLRP3 in aortic endothelial tissue of the two groups.Rat aortic endothelial cells were divided into normal control group(isolated and cultured from normal rats),positive control group(from model group),Endocan virus group and Endocan-shRNA virus group.The tubule formation ability of each group was detected by tubule formation assay.Results Serum levels of TC,TG,LDL,Endocan,IL-10 and IL-1 and mRNA and protein levels of Endocan and NLRP3 in aortic endothelial tissue were significantly higher in the model group than the control group(P<0.01).The Endocan virus group had more tubule formation,higher IL-6,IL-10,IL-1 and IL-18,and higher mRNA and protein levels of NLRP3 than the normal control group,positive control group and Endocan-shRNA virus group(P<0.05).The number of tube formation,and IL-6 levels,IL-10,IL-1βand IL-18 levels were significantly higher in the Endecan shRNA virus group than the normal control group(P<0.05).The expression of NLRP3 at mRNA and protein levels in endothelial cells of the Endecan shRNA virus group was notably higher than that of the normal control group and positive control group(5.55±1.19 vs 4.56±1.19,4.98±1.03;1.34±0.27 vs 0.78±0.12,1.09±0.21,P<0.05).Conclusion The overexpression of Endocan can promote the inflammatory injury of aortic endothelial cells,activate vascular remodeling,and is related to the expression of NLRP3 signaling pathway.
