玉米成熟胚高效苗再生体系的建立

摘  要:本研究以6种玉米自交系的成熟胚为外植体,通过优化玉米成熟胚愈伤组织诱导、继代及分化等培养环节,建立了高效的苗再生体系。结果表明,使用切除胚芽和胚根的半胚为外植体,以MS(Murashige&Skoog)培养基作为基础培养基诱导形成的愈伤组织最好,初级愈伤组织率高达80%。在继代培养过程中添加0.5 mg/mL 2,4-D和18 mg/mL Dicamba有利于提高胚性愈伤组织诱导率,将胚性愈伤组织诱导率提高到88.89%。添加20 g/L甘露醇可以将褐化率降低到19.44%。分化培养过程中加入2 mg/L Zeatin和6 mg/L TDZ可以将愈伤组织的绿苗分化率提高到48.88%。In this study,mature embryos of six maize inbred lines were used as explants,and an efficient regeneration system for maize mature embryos was established by optimizing different factors on callus induction,subculture and differentiation stages.The results showed that mature embryo with excised germinal sheath and radicle was the best explants to inoculating into MS medium,and a large number of high-quality callus could be induced,during which the induction frequency of primary callus was over 80%.The addition of 0.5 mg/mL 2,4-D and 18 mg/mL Dicamba in the subculture was beneficial to improve the induction rate of embryonic callus,and the induction rate of embryonic callus was successfully increased to 88.89%.The addition of 20 g/L mannitol was supported a decreased frequency of browning to 19.44%.Adding 2 mg/L Zeatin and 6 mg/L TDZ in the process of differentiation culture could effectively increase the differentiation rate of green seedlings to 48.88%.
