摘  要:挖掘云南文山三七内生放线菌菌种资源以及对分离到的放线菌菌株进行三七根腐病病原菌的抗菌活性评价,为生物防治三七根腐病的发生、传播、有效治理及解决三七连作障碍和后续研究三七内生放线菌的代谢产物提供菌种资源。采用三种分离方法、六种分离培养基对文山两个产地三七全株植物进行内生放线菌分离,分离到的菌株通过16S rRNA基因扩增测序鉴定到属,通过琼脂扩散法对菌株发酵液进行抗菌活性初筛。共分离到56株内生放线菌,经形态特征初步排重后,选22株代表菌株进行16S rRNA基因扩增测序,并鉴定到属,其分属于链霉菌属(Streptomyces)、放线产孢菌属(Actinomycetospora)、拟诺卡氏菌属(Nocardiopsis)、黄英菌属(Yinghuangia)四个属。对23株菌株发酵液进行抗菌活性初筛,14株菌株(60.87%)具有不同程度的抗菌活性,其中菌株S004、S006、S014、S016、S022、S042、S047、S053对三七根腐病病原菌有较好的抗菌活性,具备进一步深入研究的价值。本研究初步探索出适合三七内生放线菌分离的分离方法,获得一批三七内生放线菌菌种资源,并筛选出8株对三七根腐病病原菌有较好抗菌活性的菌株。Excavating the resources of endophytic actinomycetes strains from sanqi(Panax notoginseng)in Wenshan,Yunnan,as well as evaluating the antagonistic activity against pathogens of root rot of sanqi,these results will provide the strain resources for biological control of root rot of sanqi,effective control and solution of continuous cropping obstacle of sanqi and the follow up study of metabolic products of endophytic actinomycetes of sanqi.Three isolation methods and six isolation media were adopted to isolate endophytic actinomycetes of sanqi from two places of production in Wenshan.The isolated strains were identified to the genus by 16S rRNA gene sequences,adopting agar diffusion method to screen the antibacterial activity of the fermentation broth of the strains.The results showed that a total of 56 endophytic actinomycetes were isolated,and after rearranged morpho-charactersitically 22 representative strains were identified on the basis of 16S rRNA gene amplification and phylogenetic analysis.These strains belonged to 4 genera:Streptomyces,Actinomycetospora,Nocardiopsis,Yinghuangia.Initial antagonistic activity test of 23 strains′fermentation broth showed that 14 strains(60.87%)of them had different degree of antagonistic activities,among them strains S004,S006,S014,S016,S022,S042,S047,and S053 had better antibacterial activity against pathogens of root rot of sanqi,and having the value for further deepening study.A suitable method for isolation of endophytic actinomycetes from sanqi was preliminarily explored,and a batch of endophytic actinomycetes from sanqi was obtained,and eight strains having better antagonistic activities against sanqi root rot pathogens were screened.
