摘 要:目的研究苦参碱调控eIF4E因子对异丙肾上腺素致大鼠急性心肌肥厚的影响。方法将42只8周龄的雄性SD大鼠随机分为7组,即正常对照组(Control,0.9%NaCl)、苦参碱组(Mt,200 mg/kg)、异丙肾上腺素组(ISO,85 mg/kg)、普萘洛尔组(Propranolol,10 mg/kg)、50 mg/kg苦参碱+85 mg/kg异丙肾上腺素组(50 mg/kg Mt+ISO)、100 mg/kg苦参碱+85 mg/kg异丙肾上腺素组(100 mg/kg Mt+ISO)、200 mg/kg苦参碱+85 mg/kg异丙肾上腺素组(200 mg/kg Mt+ISO),连续给药2d。应用RT-PCR和Western-blot测定eIF4E因子的mRNA和蛋白表达水平。结果50 mg/kg Mt+ISO组、100 mg/kg Mt+ISO组和200 mg/kg Mt+ISO组的eIF4E因子蛋白表达水平明显低于ISO组(P<0.05),ISO组mRNA的表达水平是100 mg/kg Mt+ISO组的6.38倍,200 mg/kg Mt+ISO组与Control组的mRNA表达水平相近。结论苦参碱通过调控eIF4E因子的mRNA和蛋白表达水平,对大鼠的急性心肌肥厚产生保护作用。Objective To explore the effect of matrinevia regulating the eIF4E on acute myocardial hypertrophy in rats.Methods 42 male Sprague-Dawley rats were randomly divided into seven groups(n=7),control group(Control,0.9%NaCl),matrine group(Mt,200 mg/kg),isoproterenol group(ISO,85 mg/kg),propranolol group(Propranolol,10 mg/kg),matrine(50 mg/kg)+isoproterenol(85 mg/kg)group(50mg/kg Mt+ISO),matrine(100 mg/kg)+isoproterenol(85 mg/kg)group(100 mg/kg Mt+ISO),matrine(200 mg/kg)+isoproterenol(85 mg/kg)group(200 mg/kg MT+ISO).The rats were all administered for two consecutive days.RT-PCR and Western-blot were used to determine the mRNA and protein expression level of eIF4E.Results The expressions of eIF4E in the 50 mg/kg Mt+ISO group,100 mg/kg Mt+ISO group and 200 mg/kg Mt+ISO group were significantly lower than the ISO group(P<0.05).The expression of eIF4 EmRNA in ISO group was 6.38 times higher than that in the 100 mg/kg Mt+ISO group.Meanwhile,the expression of eIF4EmRNA in 200 mg/kg Mt+ISO group was close to that in the control group.Conclusion Matrine can protect rats with acute myocardial hypertrophy by regulating the mRNA and protein expression levels of eIF4E.
