摘 要:目的探究蛇床子素对柯萨奇B3病毒(CVB3)诱导的病毒性心肌炎(VMC)小鼠炎症反应的抑制作用。方法选择SPF级雄性BALB/c小鼠30只,随机分为蛇床子素治疗组(观察组)、模型组及对照组,各10只。向观察组、模型组小鼠腹腔注射CVB3病毒稀释液0.2 mL构建VMC动物模型。观察组小鼠给予50 mg·kg^(-1)·d^(-1)蛇床子素腹腔注射治疗。7 d后处死所有小鼠,留取血清和心脏标本。采用荧光定量PCR法检测心肌组织内CVB3相对表达量以及肿瘤坏死因子-α(TNF-α)和白细胞介素-1β(IL-1β)mRNA表达水平。采用酶联免疫吸附测定法检测血清中炎症因子TNF-α和IL-1β水平。采用Western blot法检测心肌组织内TNF-α和IL-1β蛋白表达水平以及蛋白激酶B(Akt)和核因子-κB(NF-κB)蛋白磷酸化水平。结果模型组小鼠心肌组织内CVB3 mRNA相对表达量、IL-1β和TNF-αmRNA表达水平高于对照组,观察组上述指标低于模型组,差异均有统计学意义(均P<0.05)。模型组小鼠血清中的IL-1β、TNF-α表达水平高于对照组,观察组小鼠血清中上述指标低于模型组,差异均有统计学意义(均P<0.05)。模型组小鼠心肌组织内Akt和NF-κB蛋白磷酸化水平、IL-1β和TNF-α蛋白表达水平高于对照组,观察组小鼠心肌组织内上述指标低于模型组,差异均有统计学意义(均P<0.05)。结论蛇床子素可能通过调控Akt/NF-κB信号通路来抑制VMC小鼠体内的炎症反应。Objective To investigate the effect of osthole on inhibiting the inflammation process of mice with viral myocarditis induced by Coxsackievirus B3(CVB3).Methods A total of 30 male BALB/c mice were randomly divided into three groups:osthole-treated group(observation group),model group and normal group(control group),with 10 mice in each group.Viral myocarditis models were established by intraperitoneal injection of 0.2 mL CVB3 in the observation group and model group.The observation group was treated with intraperitoneal injection of 50 mg·kg^(-1)·d^(-1) osthole.All mice were sacrificed 7 days later and both the serum and heart samples of the mice were collected.Myocardial CVB3 titer and TNF-αand IL-1βmRNA were detected by fluorescent quantitative PCR.Serum TNF-αand IL-1βwere detected by ELISA.Myocardial TNF-αand IL-1βprotein and phosphorylation of Akt and NF-κB protein were detected by Western blot.Results Myocardial CVB3 titer and TNF-αand IL-1βmRNA were significantly higher in the model group than in the control group,and significantly lower in the observation group than in the model group(all P>0.05).Serum TNF-αand IL-1βwere significantly higher in the model group than in the control group,and significantly lower in the observation group than in the model group(all P<0.05).Myocardial NF-κB and Akt phosphorylation and TNF-αand IL-1βprotein were significantly higher in the model group than in the control group,and significantly lower in the observation group than in the model group(all P<0.05).Conclusion Osthole may inhibit the inflammation response of mice with viral myocarditis by regulating Akt/NF-κB pathway.
