摘  要:为了深入挖掘MYB115转录因子的生物学功能,本研究以球茎甘蓝为试验材料,采用基因克隆获得球茎甘蓝MYB115转录因子,并对其进行生物信息学和荧光定量分析。生物信息学分析结果显示,BocMYB115转录因子gDNA序列全长742 bp,开放阅读框(ORF)长度为549 bp,编码182个氨基酸,具有一个高度保守的SANT结构域,属于1R-MYB类转录因子,与甘蓝型油菜的MYB115-like亲缘关系最近。荧光定量PCR结果显示,BocMYB115在绿色球茎甘蓝中的表达量显著高于紫色球茎甘蓝。本研究可为后续MYB115转录因子功能鉴定提供理论依据。Brassica oleracea L.var.caulorapa is taken as the material to further explore the biological functions of MYB115 transcription factor.Its MYB115 transcription factor is obtained by gene cloning technology.Its bioinformatics and fluorescence quantitative analysis are carried out as well.The results of bioinformatics analysis show that BocMYB115 gene sequence is 742 bp in length.ORF is 549 bp in length,encoding 182 amino acids.It has a highly conserved SANT domain belonging to 1R-MYB transcription factor,which is closely related to MYB115-like in Brassica napus.Accor-ding to the results of real-time PCR,the expression of BocMYB115 in green Brassica oleracea L.var.caulorapa is significantly higher than that in purple ones.This study lays a foundation for the functional identification of MYB115 transcription factor.
