摘 要:目的 测定三七多糖含量,观察三七多糖对Aβ42转基因果蝇影响,探讨三七多糖对阿尔茨海默症的治疗作用及其作用机制。方法 利用苯酚硫酸法测定三七总多糖含量;野生型果蝇为空白组、Aβ42转基因果蝇随机分为3组,即模型组、三七多糖组、阳性对照组(盐酸多奈哌齐),分别设置0.008、0.04、0.20、1.00、5.00 mg/mL5个给药浓度。检测各组果蝇攀爬能力;比较各组果蝇存活时间,包括平均寿命、半数死亡时间、最高寿命,并利用酶联免疫试剂盒测定果蝇脑组织中β-淀粉样斑块、丙二醛、环磷酸腺苷、乙酰胆碱酯酶含量及超氧化物歧化酶活性。结果 果蝇的攀爬指数在1 mg/mL前呈剂量依赖性,即随着给药浓度的升高,果蝇的攀爬指数相应升高(P<0.01),在给药浓度为1 mg/mL时,攀爬指数为85.11%达到峰值;三七多糖组果蝇的平均寿命、半数死亡时间、最高寿命均显著高于模型组(P<0.01);三七多糖组果蝇脑组织超氧化物歧化酶活性显著高于模型组(P<0.01);三七多糖组果蝇脑部Aβ-42(P<0.05)、丙二醛(P<0.01)、乙酰胆碱酯酶(P<0.05)含量均显著低于模型组。结论 三七多糖对Aβ42转基因果蝇具有较好的治疗作用,推测其治疗作用可能与改善β淀粉样蛋白沉积、抗氧化、减少乙酰胆碱水解等相关。Objective To determine the content of Panax notoginseng polysaccharide,to observe the effect of Panax notoginseng polysaccharide on Aβ42 transgenic Drosophila,and to investigate the therapeutic effect of it on Alzheimer’s disease and its mechanism of action.Methods The total polysaccharide content of Panax notoginseng was determined by phenol sulfuric acid method;wild-type Drosophila were divided into three groups randomly,namely,model group,Panax notoginseng polysaccharide group,and positive control group(donepezil hydrochloride).The survival time including mean life span,half death time,and maximum life span of each group were compared,and the contents of Aβ-42,MDA,cAMP,AchE,and SOD activity in the brain tissue of Drosophila were measured using ELISA kits.Results The climbing index of Drosophila was dose-dependent before 1 mg/mL,the climbing index of Drosophila increased with the increase of drug concentration(P<0.01),and peaked at 85.11%at the drug concentration of 1 mg/mL;the mean life span,time to half death and maximum life span of Drosophila in the Panax notoginseng polysaccharide group were significantly higher than those in the model group(P<0.01)The mean life span,half time to death and maximum life span of Drosophila in the Panax notoginseng polysaccharide group were significantly higher than those in the model group(P<0.01);the SOD activity in brain tissue of Drosophila in the Panax ginseng polysaccharide group were significantly higher than those in the model group(P<0.01);the content of Aβ-42(P<0.05),MDA(P<0.01)and AchE(P<0.05)in the brain of Drosophila in the Panax notoginseng polysaccharide group were significantly lower than those in the model group.Conclusion Panax notoginseng polysaccharide has better therapeutic effects on Aβ42 transgenic Drosophila,and it is speculated that its therapeutic effects may be related to the improvement ofβ-amyloid deposition,antioxidant,and reduction of acetylcholine hydrolysis.
