摘 要:目的:应用网络药理学研究熏洗方医治肛周脓肿术后创面愈合的物质基础及其可能的作用机制。方法:应用中药系统药理学数据库与分析平台(TCMSP)寻找熏洗方的主要活性成分及其药物靶点,通过基因数据库(Gene Cards)挑选相应的疾病靶点。将熏洗方的药物靶点与疾病靶点相匹配,所得交集靶点导入String数据库后构建蛋白质-蛋白质相互作用网络,使用Cytoscape 3.9.1插件计算节点信息,经拓扑分析筛选出关键靶点。使用Cytoscape 3.9.1软件构建可视化“熏洗方组成-主要活性成分-共同靶点-疾病”网络并进行分析。基于Metascape数据库对靶点进行基因生物过程分析和通路富集分析。结果:熏洗方的潜在活性药物成分有286个,包括槲皮素、芹黄素、木犀草素、山柰酚、汉黄芩素等。药物与疾病的交集靶点有164个,治疗肛周脓肿术后创面愈合的关键靶点为肿瘤蛋白p53(Tumor Protein p53,TP53)、丝氨酸/苏氨酸蛋白激酶1(Akt Serine/Threonine Kinase 1,AKT1)、丝裂原活化蛋白激酶(Mitogen-Activated Protein Kinase,MAPK)1、RELA癌基因(RELA ProtoOncogene,RELA)、肿瘤坏死因子(Tumor Necrosis Factor,TNF)、白细胞介素(Interleukin,IL)-6、雌激素受体(Estrogen Receptor,ESR)1等。主要发挥作用的信号通路为癌症通路、脂质和动脉粥样硬化、磷脂酰肌醇3激酶(Phosphatidylinositol3-kinase,PI3K)-蛋白激酶B(Akt)信号通路、MAPK信号通路、糖尿病并发症中的晚期糖基化终末产物-晚期糖基化终末产物受体(Advanced Glycation End-Products-Receptor Advanced Glycation End Products,AGE-RAGE)信号通路、雌激素信号通路等。分子对接结果表明活性成分木犀草素与关键靶点AKT1的对接结合能最低,亲和力最好。结论:熏洗方通过多途径、多靶点促进肛周脓肿术后创面愈合。PI3K-Akt信号通路、MAPK-细胞外信号调节激酶(Extracellular Signal Regulated Kinase,Erk)信号通路和雌激素信号通路可Objective:To explore action mechanism of fumigation prescription in the treatment of postoperative wound healing of perianal abscess based on network pharmacology.Methods:TCMSP database was used to find the main active ingredients and their drug targets of fumigation prescription,and the corresponding disease targets were selected through the GeneCards database.The drug targets of the fumigation prescription were matched with the disease targets,and the obtained intersection targets were imported into the String database to construct a PPI network,and the Cytoscape 3.9.1 was used to calculate the node information,and the key targets were screened out through topological analysis.Cytoscape 3.9.1 was used to construct and analyze the visual network of composition of fumigation prescription-main active components-common targets-disease.Targets were subjected to perform a gene biological process analysis and pathway enrichment analysis by Metascape database.Results:There were 286 potential active pharmaceutical ingredients in the fumigation prescription,including quercetin,apigenin,luteolin,kaempferol,and wogonin.There were 164 intersection targets between drugs and diseases,and the key targets for the treatment of would healing after perianal abscess were TP53,AKT1,MAPK1,RELA,TNF,IL-6,ESR1,etc..The main signaling pathways were cancer pathway,lipid and atherosclerosis,PI3K-Akt signaling pathway,MAPK signaling pathway,AGE-RAGE in diabetic complications signaling pathway,estrogen signaling pathway,etc..The results of molecular docking showed that luteolin had the lowest binding energy and the best affinity to the key target AKT1.Conclusion:Fumigation prescription promotes postoperative wound healing of perianal abscess through multi-channel and multi-target.The PI3K-Akt signaling pathway,the MAPK-Erk signaling pathway and the estrogen signaling pathway may be the key pathways in treating wound healing after perianal abscess with fumigation prescription.
