摘 要:目的:分析lncRNA MAFG-AS1、miR-21在宫颈癌细胞增殖与凋亡中的作用。方法:检测正常宫颈组织、宫颈癌组织与宫颈癌Hela细胞株中miR-21与lncRNA MAFG-AS1表达水平。si-MAFG-AS1、AS-miR-21转染宫颈癌Hela细胞,检测细胞活力、细胞凋亡、凋亡相关蛋白和增殖相关蛋白的表达;双荧光素酶法检验miR-21与MAFG-ASI的靶向作用关系。结果:宫颈癌组织和Hela细胞中lncRNA MAFG-AS1与miR-21表达高于正常宫颈组织和正常宫颈上皮细胞(均P<0.01)。与si-NC组相比,si-MAFG-AS1组Cyclin D1、Bcl-2蛋白表达降低,而p21、p27、Cleaved Caspase-3、Bax蛋白表达升高(均P<0.01)。与CTRL oligo组相比,AS-miR-21组Cyclin D1、Bcl-2蛋白表达减少,p21、p27、Cleaved Caspase-3、Bax蛋白表达增加(均P<0.01)。双荧光素酶检测显示,lncRNA MAFG-AS1可靶向调控miR-21。结论:LncRNA MAFG-AS1可通过靶向调控miR-21调节宫颈癌细胞的增殖与凋亡。Objective: To investigate the effects of lncRNA MAFG-AS1 and miR-21 on proliferation and apoptosis of cervical cancer cells. Methods: The expression of miR-21 and lncRNA MAFG-AS1 in normal cervical tissue, cervical cancer tissue and Hela cell line were detected. Hela cells were transfected with si-MAFG-AS1 or AS-miR-21. Then the cell viability, cell apoptosis and the expression of apoptosis related protein as well as proliferation related protein were evaluated. The target relationship between miR-21 and lncRNA MAFG-AS1 was measured using double luciferase assay. Results: LncRNA MAFG-AS1 and miR-21 were both increased in cervical cancer tissue and Hela cells, when compared with normal cervical tissue and normal cervical epithelial cells, respectively(both P<0.01). Compared to the si-NC group, Cyclin D1 and Bcl-2 were decreased, while the expression of p21, p27, Cleaved Caspase-3 and Bax were increased in the si-MAFG-AS1 group(all P<0.01). Compared with the CTRL Oligo group, Cyclin D1 and Bcl-2 were reduced, while the levels of p21, p27, Cleaved Caspase-3 and Bax were elevated in the AS-miR-21 group(all P<0.01). The results of dual luciferase assay suggested there was targeting relationship between lncRNA MAFG-AS1 and miR-21. Conclusion: LncRNA MFG-AS1 participates in regulating the proliferation and apoptosis of cervical cancer cells by targeting miR-21.
