摘 要:为进一步探讨鳖甲水提物和醇提物对人小细胞肺癌H446及人大细胞肺癌H460细胞株的体外抑制作用。通过细胞增殖实验,研究鳖甲水提物和醇提物对人小细胞肺癌H446细胞株及大细胞肺癌H460细胞株的抑制效果。首先制备鳖甲水提物与醇提物冻干粉,其次体外培养H446、H460和BEAS-2B细胞株,将其分为空白对照组及药物组(H446细胞株:2、4、8、16、32 mg/mL;H460细胞株:8、16、32、64 mg/mL;BEAS-2B细胞株:2、4、8、16、32、64 mg/mL),最后计算鳖甲水提物和醇提物对肺癌细胞H446及H460细胞株以及对肺上皮细胞的细胞存活率。研究表明:鳖甲水提物随着浓度的提高,对人小细胞肺癌H446细胞株及人大细胞肺癌H460细胞株的抑制作用逐渐增强,其在32 mg/mL时对H446细胞株的抑制效果最佳,在64 mg/mL时对H460细胞株的抑制作用最好;鳖甲醇提物随着浓度的增加,对人小细胞肺癌H446细胞株及人大细胞肺癌H460细胞株的增殖抑制作用也逐渐增强,且对H446细胞株的最佳抑制浓度在32 mg/mL,对H460细胞株的最适抑制浓度为64 mg/mL。结果表明,鳖甲水提物和醇提物对人小细胞肺癌H446细胞株及人大细胞肺癌H460细胞株均具有抑制作用,并且随着提取物浓度的升高,对两株细胞的抑制效果也更加明显。结合鳖甲提取物对正常肺上皮细胞的影响,鳖甲水提物较鳖甲醇提物抑制效果更好。In order to further investigate the in vitro inhibitory effects of water extract and alcohol extract of turtle shell on human small cell lung cancer H446 cell line and human large cell lung cancer H460 cell line,this paper studied the inhibitory e匸fects of water extract and alcohol extract of turtle shell on human small cell lung cancer H446 cell line and large cell lung cancer H460 cell line through cell proliferation experiments.First,a lyophilizing powder of water extract and alcohol extract of turtle shell was prepared.Secondly,H446,H460 and BEAS-2B cell lines were cultured in vitro and divided into blank control group and drug group(H446 cell lines:2,4,&16,32 mg/mL;H460 cell line:&16,32,64 mg/mL;BEAS-2B cell lines:2,4,8,16,32,64 mg/mL.Finally,cell survival rates were calculated for water extract and alcohol extract against lung cancer cell lines H446 and H460 and lung epithelial cells.The study showed that the water extract of turtle shell increased with the increase of concentration.The inhibitory efiect on the proliferation of human small cell lung cancer H446 cell line and human large cell lung cancer H460 cell line was gradually enhanced.The best inhibitory effect was obtained at 32 mg/mL for H446 cell line and 64 mg/mL for H460 cell line.The methanol extract of soft-shelled turtle with the increase of concentration,the inhibitory effect on the proliferation of human small cell lung cancer H446 cell line and human large cell lung cancer H460 cell line was gradually enhanced.And the best inhibitory concentration for H446 cell line is 32 mg/mL,The optimal inhibitory concentration of H460 cell line was 64 mg/mL.The results showed that the water and alcohol extracts of turtle shell had inhibitory effects on human small cell lung cancer H446 cell line and human large cell lung cancer H460 cell line.And with the increase of the extract concentration,the inhibitory effect on the two cell lines was also more obvious.Combined with the effects of turtle shell extract on normal lung epithelial cells,turtle shell
