摘  要:为筛选对人非小细胞肺癌A549、NCI-H460细胞株的增殖抑制作用较好的中药单体。通过细胞增殖实验,研究所选三种中药单体(决明蒽醌、木犀草素和去氧紫草素)对人非小细胞肺癌A549、NCI-H460细胞株的抑制效果。首先体外培养A549、NCI-H460和BEAS-2B细胞株;其次按照设定的各种中药单体的实验分组配制含药培养基;然后进行MTT实验和细胞毒理实验;最后分别计算三种单体对A549、NCI-H460和BEAS-2B细胞的细胞存活率。MTT结果显示,对A549细胞有抑制作用的中药单体为去氧紫草素和木犀草素,细胞存活率为59%和63%,决明蒽醌对A549细胞基本无抑制作用;三种单体对NCI-H460细胞均有抑制作用,细胞存活率依次为29%、50%和65%。MTT结果表明,对A549和NCI-H460细胞都有增殖抑制作用的是去氧紫草素和木犀草素,其中去氧紫草素对A549和NCI-H460细胞的增殖抑制作用优于木犀草素,且细胞存活率均在8μM时达到最低,但该浓度下BEAS-2B细胞存活率仅有64%,木犀草素对A549和NCI-H460细胞在80μM时的增殖抑制率虽低于去氧紫草素(37%vs40%,50%vs71%),但其对BEAS-2B细胞无毒性,因此,综合MTT实验和细胞毒理实验结果考虑,三种单体中木犀草素的效果最佳。In order to screen the Chinese herbal monomers with better inhibitory effects on the proliferation of human non-small cell lung cancer cell lines A549 and NCI-H460,this paper studied the inhibitory effects of three Chinese herbal monomers(Obtusiflin,Luteolin and Deoxyshikonin)on human non-small cell lung cancer cell lines A549 and NCI-H460 through cell proliferation experiment.Firstly,A549,NCI-H460 and BEAS-2B cell lines were cultured in vitro.Secondly,phannaceutical medium was prepared according to the experimental groups of three traditional Chinese medicine monomers.Then MTT assay,cyto toxicity assay and cell morphology observation were performed.Finally,the cell viability of A549,NCI-H460 and BEAS-2B cells were calculated.MTT results showed that the Chinese herbal monomers with inhibitory effects on A549 cells were Deoxy-shikonin and Luteolin,and the cell survival rate was 59%and 63%,while the Obtusiflin had no inhibitory effect on A549 cells.All the three monomers inhibited NCI-H460 cells,and the cell survival rate was 29%,50%and 65%.MTT results showed that Deoxyshikonin and Luteolin had proliferation inhibition on both A549 and NCI-H460 cells.Deoxyshikonin had better proliferation inhibition on A549 and NCI-H460 cells than Luteolin,and the cell survival rate reached the lowest at 8 piM.However,at this concentration,the survival rate of BEAS-2B cells was only 64%.Although the inhibitory rate of Luteolin on the proliferation of A549 and NCI-H460 cells at 80 p,M was slightly lower than that of Deoxyshikonin(37%vs40%,50%vs71%),it has no toxicity to BEAS-2B cells.Therefore,considering the results of MTT assay and cytotoxicity assay,Luteolin has the best effect among the three monomers.
