摘  要:为了明确灰葡萄孢过氧化氢酶BcCAT2在病菌生长发育时期和侵染过程中的功能,本研究利用生物信息学方法,对BcCAT2及其编码蛋白进行分析;构建了BcCAT2敲除载体,利用原生质体转化方法,获得了BcCAT2敲除突变体;利用PCR和qRT-PCR技术,对所获得的转化子进行鉴定;对野生型菌株和△Bccat2的表型(菌落形态、菌丝形态、生长速度、产孢量等)和致病力进行分析。结果发现,BcCAT2基因的缺失会导致菌丝形态纤细、生长速度缓慢、几乎不产菌核和分生孢子,且致病力明显下降,表明BcCAT2基因正调控病菌的生长发育和致病力。研究结果为进一步阐明灰葡萄孢过氧化氢酶BcCAT2基因调控病菌生长发育和致病力的分子机制奠定了基础。In order to clarify the function of catalase BcCAT2 in the growth and development stage and infection process of Botrytis cinerea,this study used bioinfonnatics methods to analyze BcCAT2 and its coding protein.BcCAT2 knockout vector was constructed to create BcCAT2 knockout mutant by protoplast transformation.PCR and qRT-PCR were used to identify the transformants.The phenotypes(colony morphology,mycelium morphology,growth rate,spore yield,etc.)and pathogenicity of the mutant h.Bccat2 and wild type strains were analyzed.The results showed that the deletion of BcCAT2 gene resulted in thin mycelia,slow growth,few sclerotia and conidia and decreased pathogenicity,indicating that BcCAT2 gene regulate the growth,development and pathogenicity of bacteria.The results lay a foundation for further elucidating the molecular mechanism of Botrytis cinerea BcCAT2 gene regulating the growth,development and pathogenicity of the pathogen.
