摘  要:为探讨鼠李糖乳杆菌GR-1(Lactobacillus rhamnosus GR-1,L.rhamnosus GR-1)对大肠杆菌(Escherichia coli)诱导的奶牛子宫内膜上皮细胞(Bovineendometrialepithelialcells,BEECs)炎性损伤的保护作用及其机制,本研究建立BEECs细胞炎性损伤模型,利用Real-time qPCR和Western blot技术,观察在BAY11-7082或siRNA干预前后,L.rhamnosus GR-1对E.coli触发的信号通路及炎性因子表达的影响。结果显示,L.rhamnosusGR-1可以显著抑制由E.coli引起的NF-κB和TRIF通路的激活及炎性因子的表达,在BAY11-7082干预后,E.coli诱导的NF-κB通路的激活及炎性因子的表达得到了显著的抑制。同样,在添加TRIF siRNA后,E.coli诱导的TRIF mRNA及炎性因子的表达水平也显著下降,产生和L.rhamnosus GR-1一致的抗炎效果。本研究表明L.rhamnosus GR-1可通过介导NF-κB和TRIF通路调节E.coli诱导的BEECs的炎性损伤,为奶牛子宫内膜炎的防治提供有益参考。The purpose of this study was to investigate the protective effect and mechanism of Lactobacillus rhamnosus GR-1(L.rhamnosus GR-1)on inflammatory injury of bovine endometrial epithelial cells(BEECs)induced by E.coli.In this study,the model of inflammatory injury of BEECs was established.Real-time qPCR and western blot were used to observe the effects of L.rhamnosus GR-1 on the signal pathway triggered by E.coli and the expression of inflammatory cytokines before and after the intervention of BAY11-7082 or siRNA.The results showed that L.rhamnosus GR-1 significantly inhibited the activation of NF-kB and TRIF pathways and the expression of inflammatory cytokines was induced by E.coli.After the intervention of BAY 11-7082,the NF-kB pathway was activated by E.coli and the expression of inflammatory cytokines were significantly inhibited.Similarly,after the addition of TRIF siRNA,the mRNA expression of TRIF that was induced by E.coli decreased significantly resulting in an anti-inflammatory effect which was consistent to that of L.rhamnosus GR-1.This study showed that L.rhamnosus GR-1 can reduce the inflammatory damage of BEECs induced by E.coli by mediating NF-kB and TRIF pathways,which provided a useful reference for the prevention and treatment of endometritis in dairy cows.
