摘  要:羧甲基纤维素钠(CMC-Na)是一种食品添加剂,常用作增稠剂、稳定剂等.采用荧光光谱法(FS)和紫外-可见光谱法(UV-Vis)研究了CMC-Na与牛血清白蛋白(BSA)之间的结合机制.研究结果表明:CMC-Na对BSA猝灭类型为动静态联合猝灭;得到的结合常数表明CMC-Na与BSA具有较强的结合能力;得到的结合位点数表明CMC-Na在BSA上存在1个结合位点;得到的3个不同温度(293 K、298 K、303 K)下的热力学参数ΔS^(θ)、ΔH^(θ)和ΔG^(θ)均小于零表明CMC-Na与BSA的结合过程是自发的;作用力类型研究结果表明CMC-Na与BSA之间的作用力主要为范德华力;紫外-可见光谱研究结果表明,CMC-Na诱导BSA的构象发生了变化.Sodium carboxymethylcellulose(CMC-Na)was a kind of food additive,which was often used as thickening agent,emulsion stabilizer,etc.The binding mechanism between bovine serum albumin(BSA)and CMC-Na by fluorescent spectrometry(FS)and ultraviolet-visible absorption spectrometry(UV-Vis)were studied.The results showed that the type of CMC-Na’s quenching of BSA is dynamic and static combined quenching.The obtained binding constants indicated that CMC-Na had strong binding ability with BSA.The number of binding sites obtained indicated that CMC-Na had one binding site on BSA.ΔS^(θ),ΔH^(θ),andΔG^(θ),the thermodynamic parameters at three different temperatures(293K,298 K,and 303K)were all less than zero.It indicated that the binding process of CMC-Na and BSA is spontaneous and driven by enthalpy and the main driving forces are hydrogen bond and Van der Waals’force.Besides,the research results of UV-Vis absorption spectrometry showed that CMC-Na induced the changes in the conformation of BSA.
