摘 要:【目的】筛选对蒙古拟地甲(Gonocephalum mongolicum)成虫具高毒力的球孢白僵菌(Beauveria bassiana)菌株,为此害虫的生物防治提供真菌资源。【方法】将5株供试球孢白僵菌配制为1.0×10^(8)孢子/mL的悬浮液,采用浸虫法测定对蒙古拟地甲成虫的致死率以及致死中时(LT50)。筛选出高毒力菌株后,测定该菌株对试虫的致死中浓度(LC50)。同时利用体视镜观察试虫被该菌株侵染后的症状。【结果】在5株供试菌株中,Bb06对蒙古拟地甲成虫致病力最高。该菌株处理20 d后引起的试虫校正死亡率为82.97%,显著高于其他菌株;其LT50为11.09 d,显著短于其他菌株。计算结果表明Bb06处理20 d后对试虫的LC50为0.40×10^(6)孢子/mL。蒙古拟地甲成虫被Bb06侵染后活动逐渐变得迟缓且进食量减少,试虫头部以及腹部陆续长出白色菌丝,直至将整个虫体覆盖,并产生大量分生孢子。【结论】Bb06菌株对蒙古拟地甲成虫致病力较高,具有进一步开发利用的价值。[Objective]This study aims to screen Beauveria bassiana strains with high virulence to the adults of Gonocephalum mongolicum,thus providing fungal resources for the biological control of the pest species.[Method]The conidial suspensions of five B.bassiana strains were prepared at a concentration of 1×10^(8)conidia/mL.The mortality rate and median lethal time(LT50)of G.mongolicum adults infected by fungal suspensions were determined by the immersion method.After selection of a fungal strain with highest virulence to G.mongolicum,the median lethal concentration(LC50)was determined.In addition,the infection process of Bb06 and the morphological characteristics of G.mongolicum after the infection were observed by using a stereoscope.[Result]Among five B.bassiana strains,Bb06 showed the highest pathogenicity to the adults of G.mongolicum.82.97%corrected mortality was observed for individuals at 20 days after treatment with Bb06,and the mortality was significantly higher than that of other B.bassiana strains.The LT50 of individuals infected with Bb06 was 11.09 d,which significantly shorter than the time of other fungal strains.The results showed that the LC50 of Bb06 against G.mongolicum adults was 0.4×10^(6)conidia/mL at 20 days after treatment.After the infection with Bb06,diseased G.mongolicum adults appeared sluggish in their movement and the insects reduced their food intake.Subsequently,the white mycelia grew on the head and abdomen of the test individuals and covered the whole insect body,and produced a large number of conidia.[Significance]The strain Bb06 showed a high pathogenicity against the adults of G.mongolicum.Therefore,the strain has a great value of development and utilization.
