摘 要:【目的】为外生菌根真菌的开发利用提供菌种资源和理论基础。【方法】采集松科植物根部外生菌根真菌子实体,通过分离获得LS07、LS062和LS065等3株纯培养菌株,依据形态学特征和ITS序列分析对分离菌株进行鉴定,同时采用单因素试验方法,研究不同培养条件和营养成分对菌丝生长的影响。【结果】LS07菌株经鉴定为琥珀乳牛肝菌(Suillus placidus),其菌丝生长最适单糖为果糖,最适双糖为麦芽糖,最适多糖为棉子糖和果胶,最适氮源为铵态氮,最适pH为5,最适生长温度为25℃;LS062菌株经鉴定为大孢硬皮马勃(Scleroderma bovista),其菌丝生长最适单糖为果糖,最适双糖为乳糖,最适多糖为棉子糖,最适氮源为铵态氮,最适pH为5,最适生长温度为30℃;LS065菌株经鉴定为东方豆马勃(Pisolithus orientalis)。其菌丝生长最适单糖为果糖,最适双糖为蔗糖、麦芽糖,最适多糖为淀粉,最适氮源为铵态氮,最适pH为6,最适生长温度为35℃。【结论】研究表明不同菌株最适生长温度和碳源具有显著差异,为外生菌根真菌分离和扩培提供了理论依据。[Objective]This study was conducted to provide strain resources and theoretical basis for the development and utilization of ectomycorrhizal fungi.[Method]Three pure cultured fungal strains,LS07,LS062 and LS065,were isolated from fruiting bodies of ectomycorrhizal fungi collected from the roots of Pinaceae trees.The isolated strains were identified according to morphological characteristics and ITS sequence analysis.The effects of different culture conditions and nutrients on mycelial growth were studied by single factor experiment.[Result]The results showed that strain LS07 was identified as Suillus placidus.The optimum monosaccharide for mycelial growth of LS07 was fructose,the optimum disaccharide was maltose,the optimum polysaccharide was raffinose and pectin.Besides,the optimum nitrogen source for mycelial growth of LS07 was NH4+-N,the most suitable pH was 5,and the most suitable growth temperature was 25℃.The strain LS062 was identified as Scleroderma bovista.Its optimum monosaccharide,disaccharide,and polysaccharide for mycelial growth was fructose,lactose,and raffinose,respectively.The optimum nitrogen source for mycelial growth of LS062 was NH4+-N.The optimum pH was 5,and the optimum growth temperature was 30℃.The strain LS065 was identified as Pisolithus orientalis.Its optimum monosaccharide,disaccharide,and polysaccharide for mycelial growth was fructose,sucrose and maltose,and starch,respectively.The optimum nitrogen source was NH4+-N,the optimum pH was 6,with the optimum growth temperature 35℃.[Conclusion]This study indicated the optimum growth temperature and carbon source of different strains were significantly different,providing a theoretical basis for the isolation and cultivation of ectomycorrhizal fungi.
