摘 要:建立同时测定经典名方二冬汤中芒果苷、金丝桃苷、黄芩苷、汉黄芩苷、甘草酸铵5种成分的高效液相色谱(HPLC)分析方法,并测定其在15批物质基准中的含量。采用MS Target SB-C18色谱柱(250 mm×4.6 mm,5μm),以0.1%磷酸水溶液-乙腈为流动相,梯度洗脱;体积流量为1.0 mL/min;柱温30℃;检测波长为254 nm。同时,采用SPSS软件对15批物质基准中各成分的含量进行聚类分析,以评价样品间的质量差异。结果显示5种成分色谱峰分离度良好,芒果苷、金丝桃苷、黄芩苷、汉黄芩苷的质量浓度分别在2.32~23.2、2.244~22.44、16.98~169.8、3.776~37.76、2.416~24.16μg·mL^(–1)线性范围良好(r^(2)≥0.9992);平均加样回收率分别为105.78%、99.61%、103.26%、99.19%、96.61%,RSD均<3%;15批物质基准中5种成分的质量分数依次为0.09%~0.26%、0.11%~0.16%、1.58%~2.46%、0.30%~0.45%、0.15%~0.31%;聚类分析将15批物质基准聚为4类。所建立的多成分含量测定方法结果稳定、可行、方法易操作、准确,可用于经典名方二冬汤物质基准及其制剂的质量控制。A high performance liquid chromatography(HPLC)method was established for the simultaneous determination of mangiferin,hypericin,baicalin,wogonin and Ammonium Glycyrrhizinate in Erdong decoction,a classic famous prescription,and its content in 15 batches of material standards.The quantitative analysis was carried out on a Agilent SB-C18(250 mm×4.6 mm,5μm)with mobile phase of acetonitrile-0.1%phosphoric acid aqueous solution for gradient elution,the flow rate was 1.0 mL/min,the column temperature was maintained at 30℃,the detection wavelengths were set as 254 nm.At the same time,SPSS was used to cluster the contents of each component in the 15 batches of material benchmark to evaluate the quality differences between samples.The results showed that the chromatographic peaks of the five components were well separated.The linear ranges of the above 5 components were 2.32-23.2,2.244-22.44,16.98-169.8,3.776-37.76,2.416-24.16μg·mL^(–1),respectively(r^(2)≥0.9992).The average of them were 105.78%,99.61%,103.26%,99.19%,96.61%,respectively(RSD<3%).The mass fractions of 5 components in the 15 batches of material benchmark were 0.09%-0.26%,0.11%-0.16%,1.58%-2.46%,0.30%-0.45%,0.15%-0.31%;The 15 batches of material benchmarks were clustered into 4 categories by cluster analysis.The established method for simultaneous determination of multi-components is stable,reliable,simple and accurate,and can be used for the material standard of Erdong Decoction and the quality control of its preparations.
