摘 要:采用ISSR分子标记技术对5种杜鹃属植物山顶矮林优势种的遗传多样性和亲缘关系进行研究,探讨比较5种植物的遗传多样性水平和亲缘关系。结果表明,5条引物共扩增出131个位点,均为多态性位点,多态性位点比率(PPB)为100%;5种植物93个样本总的观察等位基因数(Na)、有效等位基因数(Ne)、Nei’s基因多样性指数(H)和Shannona信息指数(I)分别为2.0000、1.3243、0.2161和0.3535。变色杜鹃与猴头杜鹃的遗传相似系数最大,为0.8969。聚类分析将5种杜鹃属植物分为三个类群,变色杜鹃和猴头杜鹃为一类,井冈山杜鹃和光枝杜鹃为一类,云锦杜鹃为单独的一类,聚类分析支持变色杜鹃为猴头杜鹃的变种。5种杜鹃属植物的遗传多样性水平较低,ISSR分子标记技术可用于杜鹃属植物之间的遗传多样性和亲缘关系研究,可为杜鹃属植物的分类鉴定提供分子证据。The genetic diversity and genetic relationship of five Rhododendron montane elfin forests dominant species were studied by molecular ISSR markers,and 5 plants'genetic diversity level and genetic relationship were explored and compared.The results showed that a total of 131 loci were amplified with 5 primers,and all of them were polymorphic loci.The ratio of polymorphic loci(PPB)was 100%.The total observed allele number(Na),effective allele number(Ne),Nei’s gene diversity index(H)and Shannona information index(I)of 93 samples from 5 plant species were 2.0000,1.3243,0.2161 and 0.3535,respectively.The genetic similarity coefficient between Rhododendron simiarum Hance var.versicolor(Chun et Fang)Fang and Rhododendron simiarum Hance was the greateast of 0.8969.The cluster analysis divided five Rhododendron species into three groups:Rhododendron simiarum Hance var.versicolor(Chun et Fang)Fang and Rhododendron simiarum Hance,Rhododendron jinggangshanicum P.C.Tam and Rhododendron haofui Chun et Fang,and Rhododendron fortune Lindl.,the cluster analysis supported that Rhododendron simiarum Hance var.versicolor(Chun et Fang)Fang was a variety of Rhododendron simiarum Hance.The level of genetic diversity of five species of Rhododendron was low.ISSR molecular marker technique could be used to study the genetic diversity and genetic relationship among plants of Rhododendron,and provide molecular evidence for the classification and identification of Rhododendron.
