摘 要:以未沙藏、沙藏后萌发与未萌发的北沙参种子为实验材料,运用数字基因表达谱(DGE)技术,对种子的转录本进行功能注释和差异分析。通过分析沙藏和未沙藏以及沙藏后萌发与未萌发的北沙参种子基因表达差异,探究沙藏打破北沙参种子休眠的机理和种子萌发的分子机制。结果表明,测序后共得到218387701条原始序列,有7.85%的Unigene注释到7个数据库中。差异基因的通路分析中,沙藏与未沙藏的种子基因差别在核糖体,萌发与未萌发的种子主要区别在苯丙烷生物合成途径。北沙参种子沙藏后可以引起核糖体基因的表达,打破种子休眠。香豆素作为苯丙烷合成的重要产物,其基因的差异表达,可能与种子的萌发有密切关系。Methods digital gene expression profiling(DGE)technology was used to annotate and analyze the functional differences of Glehnia littoralis seed transcripts from the non-stratification seeds,germinated and ungerminated seeds after stratification.To explore the mechanism of stratification breaking Glehnia littoralis seed dormancy and the molecular mechanism of seed germination by analyzing the gene expression differences between stratification and non-stratification seeds,as well as germinated and non-germinated seeds after stratification.Results showed that a total of 218387701 clean reads were obtained after sequencing,and 7.85%of Unigene were annotated into 7 databases.In the pathway analysis of differentially expressed genes,the gene difference between stratification and non-stratification seeds was found in the ribosome,and the main difference between germinated and ungerminated seeds was in the phenylpropane biosynthesis pathway.The expression of ribosomal genes could be resulted and seed dormancy could be broken after stratification.Coumarin was as an important product of phenylpropane synthesis,and its gene differential expression might be closely related to seed germination.
