摘 要:本文为五味子藤茎有效成分的提取纯化工艺条件优选,对其抗氧化活性及抗UVB照射人皮肤角质细胞(HaCat)损伤细胞的作用进行研究,本研究以清除DPPH自由基能力、羟自由基能力、总抗氧化能力以及HaCat细胞增殖情况为测定指标,优选五味子藤茎最佳提取溶媒及提取纯化工艺条件。结果表明,五味子藤茎以水为溶剂进行提取,在生产成本及抗氧化活性上具有较好的优势;水提取工艺条件通过单因素考察及正交试验,确定在料液比为1:25、提取温度100℃、提取时间60 min和提取3次的条件下,五味子藤茎提取率最高;在纯化工艺条件筛选中,50%乙醇洗脱的五味子藤茎提取物对UVB照射HaCat细胞形成的氧化损伤有明显的保护作用。五味子藤茎水提物经过纯化具有较好的抗氧化活性,并对UVB照射HaCat细胞具有保护作用。The extraction and purification conditions ofthe effective components from the cane of Schisandra chinensis were optimized,and its antioxidant activity and anti UVB irradiated HaCat cells were studied.The ability of scavenging DPPH free radical,hydroxyl free radical,total antioxidant capacity and HaCat cell proliferation were the measurement indexes,and the best extraction solvent,extraction and purification process conditions were optimized.The results showed that the cane of Schisandra chinensis was extracted with water as the solvent,which had better production cost and antioxidant activity.The optimal extraction condition were as follow,solid-liquid ratio was 1:25,extraction temperature was 100℃,extraction time waw 60 min and extraction was times 3.In the screening of purification condition,the stem extract of Schisandra chinensis eluted by 50%ethanol had obvious protective effect on oxidative damage ofHaCat cells after UVB irradiation.After purification,the aqueous extract of Schisandra cane has good antioxidant activity and has a protective effect on HaCat cells irradiated by UVB.
