摘 要:建立反相高效液相色谱法(RP-HPLC)测定安石榴苷的血药物浓度,并用于大鼠灌胃后的药动学研究。采用COSMOSIL-Chol-ester C18色谱柱(4.6 mm×250 mm,5μm),以甲醇-0.1%磷酸为流动相梯度洗脱,检测波长370 nm,柱温40℃,流速1.0 mL/min。SD大鼠单次口服安石榴苷(300 mg/kg)后,眼眶静脉丛采血,检测24h内11个时间点血浆中的药物浓度。安石榴苷在2.18~58.00μg/mL范围线性关系良好,定量下限(LLOQ)2.18μg/mL,日内、日间精密度RSD<15%,提取回收率为89.67%~115.56%。PK Slutions 2.0^(TM)药动学软件拟合主要药动学参数C_(max)、T_(max)、AUC_((0-t))分别为(26.33±5.65)μg/mL、(1.67±0.52)h和(243.40±111.62)(μg·h)/mL。所建立RP-HPLC法测定安石榴苷的血药浓度简便、准确、专属性强,给大鼠灌服安石榴苷后,其吸收、分布迅速,体内药动学呈开放的二室模型。An RP-HPLC method was established to determine plasma concentration of punicalagin and study its pharmacokinetic behavior in rats after received intragastric administration.The separationwas performed onthe COSMOSIL-Cholester C18 column(4.6mm×250mm,5μm)at 40℃with a gradient elution of methanol-0.1%phosphoric acid.The detection wavelength was set at 370 nm,and the flowrate was 1.0 mL/min.SD rats were given punicalagin by intragastric administration at a dose of 300 mg/kg.The blood samples were collected fromthe orbital venous plexus and the plasma concentrations of punicalagin were deteanined at 11 time points over 24 h.The linear range of punicalagin was 2.18~58.00 g/mL(r=0.9976)with the lowest limit of quantitation(LLOQ)was 2.18 g/mL.The RSDs of the inter-day and intra-day precision were less than 15%,and the extraction recovery rates were 89.67%~115.56%.The main pharmacokinetic parameters C_(max),T_(max) and AUC_((0-t))calculated by PK Slutions 2.0TMsoftware program were(26.33±5.65)g/mL,(1.67±0.52)h and(243.40±111.62)(g·h)/mL,respectively.The RP-HPLCmethod is accurate and sensitive,which is suitable for the determination of punicalagin in rat plasma.The absorption and distribution of punicalagin in rat after intragastric administration was rapid.Its pharmacokinetics behavior was fitted to two-compartment model.
