摘  要:目的:研究芒果苷对高糖状态下MC3T3-E1成骨细胞骨向分化能力的影响。方法:采用低糖培养基(对照组)、高糖培养基(高糖组)和含40μmol/L芒果苷的高糖培养基(高糖+芒果苷组)分别体外培养MC3T3-E1成骨细胞。培养14 d后,茜素红染色法定量分析检测成骨细胞钙盐沉积量,酶联免疫吸附试验(ELISA)检测细胞成骨相关蛋白碱性磷酸酶(ALP)、骨钙蛋白(OCN)的表达水平。结果:对照组光密度(OD)值、ALP、OCN蛋白表达水平高于高糖组与高糖+芒果苷组,高糖+芒果苷组OD值、ALP、OCN蛋白表达水平高于高糖组,差异有统计学意义(P<0.05)。结论:芒果苷可以促进高糖状态下MC3T3-E1成骨细胞的骨向分化。Objective:To study the effect of mangiferin on the osteogenic differentiation ability of MC3T3-E1 osteoblasts under high glucose status.Methods:MC3T3-E1 cells were cultured in vitro with low glucose medium(control group),high glucose medium(high glucose group)and high glucose medium containing 40μmol/L mangiferin(high glucose+mangiferin group).After 14 days of culture,alizarin red staining was used to quantitatively analyze the detect the amount of calcium deposition in osteoblasts and enzyme linked immunosorbent assay(ELISA)was used to detect the expression levels of osteogenesis-related proteins alkaline phosphatase(ALP)and osteocalcin(OCN).Results:The optical density(OD)values,ALP and OCN protein expression levels in the control group were higher than those in the high glucose group and the high glucose+mangiferin group,and the OD value,ALP and OCN protein expression levels in the high glucose+mangiferin group were higher than those in the high glucose group,and the difference was statistically significant(P<0.05).Conclusion:Mangiferin can promote the osteogenic differentiation of MC3T3-E1 osteoblasts under high glucose status.
