姜酚油提取工艺优化和HPLC特征图谱的建立

摘　　要：目的 研究姜酚油的最佳提取工艺路线,并建立姜酚油的HPLC特征图谱,为姜酚油的质量控制方法建立提供依据。方法 以6-姜酚、8-姜酚、6-姜烯酚和10-姜酚为主要指标性成分,采用HPLC特征图谱结合含量测定,考察AB-8大孔吸附树脂对姜酚油的吸附性能和洗脱参数,确定最佳提取工艺。结果 最佳工艺条件为:提取溶剂选用80%乙醇,比上柱量是10 g/g(药材/干树脂),生药上样浓度0.8 g/mL,40%乙醇、80%乙醇以2 mL/min速度依次洗脱,收集80%乙醇洗脱液。通过AB-8大孔吸附树脂纯化提取的姜酚油中4种指标性成分总含量为(23.56±0.17)%,且6批姜酚油的相似度均大于92%。结论 确立的提取工艺稳定、可控,确立的高效液相特征图谱可为姜酚油的研究及应用提供参考。Objective To study the best extraction process of gingerol oil and establish the HPLC characteristic chromatogram of gingerol oil,provide basis for the establishment of quality control method of gingerol oil.Methods 6-Gingerol,8-gingerol,6-shogaol and 10-gingerol as four indication components in Gingerol oil,the adsorption performance and elution parameters of Gingerol oil in AB-8 macroporous resin were investigated by HPLC characteristic chromatogram with content determination of 6-gingerol.The optimized extraction process was determined.Results The best process conditions were 80% ethanol as extraction solvent,column loading10 g/g(crude drug/resin),drug loading concentration 0.8 g/mL,elution solvents 40% and 80% ethanol,elution speed 2 mL/min,and the 80% ethanol eluate was collected.Through purification with AB-8 macroporous adsorption resin,the total content of four indicative components was determined as(23.56±0.17)%,the similarity of 6 batches of Gingerol oil is more than 92%.Conclusion The extraction process was stable and controllable.The estab1ished characteristic chromatogram of HPLC can provide experimental reference for the research and application of Gingeroloil.
