摘  要:目的 分析广陈皮在陈化过程中主要成分的定性定量变化及褐变反应类型。方法 采用LC-MS和HPLC法对陈化过程中主要成分进行定性定量分析。采用UV和HPLC法研究自然陈化和模拟陈化广陈皮中美拉德褐变反应底物、中间产物等含量变化及抗氧化活性。结果 广陈皮中共鉴定出40个黄酮类成分和2个生物碱成分及柠檬苦素。5种黄酮类化合物含量随着陈化时间延长呈增加趋势,辛弗林含量呈下降趋势。随着陈化时间的延长,氨基酸和还原糖含量呈降低趋势,5-羟甲基糠醛含量明显增加,样品液pH下降,ABTS抗氧化活性增强。模拟陈化样品随着加热时间延长,美拉德褐变反应指标变化趋势同自然陈化样品。结论 陈化过程中发生了美拉德褐变反应,可能是导致广陈皮颜色加深的机制。5-羟甲基糠醛可作为广陈皮陈化中美拉德褐变反应的指标成分。Objective To analyze the change of compositions and Browning reaction type in Guang Citri reticulatae Pericarpium(GCRP) during ageing.Methods LC-MS and HPLC were applied to analyze the main compositions in the ageing process.UV and HPLC methods were used to study the contents of substrates,intermediates from Maillard Browning reaction and antioxidant activities of natural ageing and simulated ageing samples.Results A total of 40 flavonoids,2 alkaloids and limonin were identified in GCRP.The contents of five flavonoids increased with ageing time,while the content of synephrine decreased.With the ageing time prolonged,the contents of amino acid and reducing sugar decreased,the content of 5-hydroxymethylfurfural increased significantly,the pH of sample solution decreased,and the antioxidant activity of ABTS increased.The variation trend of Maillard Browning reaction indexes of simulated ageing samples was the same as that of natural ageing samples.Conclusion Maillard Browning reaction occurs during the ageing process of GCRP,which may be one of the mechanisms for color darkening.5-Hydroxymethylfurfural can be used as an important index composition of Maillard Browning reaction in the ageing process.
