摘  要:目的通过单因素考察和星点设计-效应面法对制备工艺进行优化。方法HPLC法测定姜黄素的含量;采用薄膜分散法制备姜黄素纳米脂质体(Cur-np);以包封率(EE)及载药量(DL)为主要指标对其进行质量评价,在单因素考察基础上,以姜黄素用量、磷酸缓冲盐溶液(PBS)的pH值、PBS浓度,进行3因素5水平的正交试验,考察对EE、DL的影响,并采用星点设计-效应面法对处方进行优化,对分析结果得出最优处方进行验证。结果Cur-np的最优处方是姜黄素用量为1mg、PBS的pH值为5.5、PBS浓度为30mmol·L-1,根据优化处方工艺制备的Cur-np包封率为93.76%,载药量为7.54%。结论根据优化处方工艺制备的Cur-np包封率载药量均较高。Objective To optimize the preparation process by single factor investigation and central composite design response surface methodology.Methods The content of curcumin was determined by HPLC;curcumin nano liposomes(Cur-np)were prepared by membrane dispersion method;the encapsulation efficiency(EE)and drug loading(DL)were used as the main indicators to evaluate their quality.On the basis of single factor investigation,the orthogonal test with three factors and five levels was conducted to investigate the effects of curcumin dosage,pH value of phosphate buffer solution(PBS),and PBS concentration on EE and DL.The prescription was optimized by star point design response surface methodology,and the analysis results were verified to obtain the optimal prescription.Results The optimal formulation of Cur-np was curcumin 1mg,PBS pH5.5 and PBS concentration 30 mmol·L-1.The encapsulation efficiency of Cur-np was 93.76%and the drug loading was 7.54%.Conclusion The encapsulation efficiency and drug loading of Cur np prepared according to the optimized formulation and technology are high.
