芪蛭胶囊对脑缺血再灌注损伤大鼠自噬相关因子LC3、p62表达水平的影响

摘　　要：目的通过研究益气活血通络中药芪蛭胶囊对脑缺血再灌注损伤大鼠自噬相关因子LC3、p62表达的影响,阐明该方拮抗脑缺血再灌注损伤的可能分子机制。方法将大鼠随机分为sham组、I/R组、3-MA组、中药组。中药及3-MA抑制剂干预后,复制MCAO/R模型,再灌注24 h后进行神经功能评分;TTC染色评定脑梗死体积;免疫荧光、Western-Blot及电镜检测自噬发生水平、自噬相关蛋白表达水平、细胞损伤程度。结果与I/R组相比,中药组大鼠神经功能损伤显著减轻,脑梗死体积显著缩小,LC3BⅡ/Ⅰ比值明显升高,p62蛋白表达量显著降低,中药组细胞形态及细胞器保留较完整,有较多自噬小体及自噬溶酶体出现。结论芪蛭胶囊可有效缓解脑梗死及神经功能损伤,提高LC3,降低p62的表达,进一步激活细胞自噬,从而有效拮抗CIRI,保护神经元。Objective To clarify the possible molecular mechanism of Qizhi Capsule's antagonism to cerebral ischemia reperfusion injury through investigating the effects of Qizhi Capsule which could replenishing Qi,activating blood and collaterals on the expression of autophagy related factors LC3 and p62 in rats with cerebral ischemia-reperfusion injury.Methods Rats were randomly divided into sham group,I/R group,3-MA group and Traditional Chinese medicine group.3-MA inhibitor and Chinese medicine intervention were carried out before MCAO/R model replication.Neurological Severity score was performed after 24h of reperfusion.After that,cerebral infarct volume was assessed by TTC staining.The degree of autophagy occurrence,the expression levels of autophagy related proteins and the condition of cells were detected by immunofluorescence,Western-Blot and electron microscopy.R esults Compared with the I/R group,the TCM group significantly reduced neurological impairment,lessened cerebral infarct volumes,raised LC3BⅡ/Ⅰratios,reduced p62 protein expression quantity.And the TCM group showed more complete cell morphology and organelle retention with more autophagosomes and autophagolysosomes appearing.Conclusion Qizhi Capsule can effectively alleviate cerebral infarction and neurological impairment,increase LC3,decrease p62 expression and further activate cell autophagy,thus effectively antagonizing CIRI and protecting neurons.
