摘 要:目的观察消饮化瘀方对CHF大鼠β肾上腺素受体信号通路的影响。方法72只SD大鼠随机分为空白组和造模组,造模组采用腹腔注射阿霉素6周复制CHF模型,随后使用彩色多普勒超声心动仪检测模型是否复制成功。模型复制成功后将大鼠随机分为模型组、中药低剂量组(7g/kg)、中药中剂量组(14g/kg)、中药高剂量组(28g/kg)及美托洛尔组(10mg/kg),分别灌胃给予相应的药物,每日一次,连续4周。Elisa检测心肌组织中脑钠肽(BNP)含量。Western Blot法检测心肌组织中β1-AR、PI3KCG、P-AKT蛋白含量,Real Time PCR检测心肌组织中β1-ARmRNA、PI3KCGmRNA表达水平。结果与空白组相比,模型组心功能降低,BNP、β1-AR、β1-ARmRNA表达明显升高,PI3KCG、P-AKT、PI3KCGmRNA表达明显降低。与模型组比较,各治疗组BNP、β1-AR、β1-ARmRNA表达明显降低,PI3KCG、PI3KCGmRNA表达明显升高;中药中剂量、中药高剂量及美托洛尔P-AKT表达明显升高。结论消饮化瘀方可能是通过β-AR信号通路,抑制细胞凋亡,降低细胞Ca 2+浓度,改善心功能,延缓心室重构。Objective To observe the effect of Xiaoyin Huayu prescription onβadrenal receptor signaling pathway in CHF rats.Methods 72 SD rats were randomly divided into blank group and model group.Model group was intraperitoneally injected adriamycin for 8 weeks to replicate CHF model,and then color Doppler echocardiography was used to detect whether the model was successfully replicated.After the model was successfully replicated,the rats were randomly divided into model group,low dose group(7g/kg),medium dose group(14g/kg),high dose group(28g/kg)and metoprolol group(10mg/kg).The rats were given corresponding drugs by intragastric administration,once a day,for 4 weeks.The content of brain natriuretic peptide(BNP)in myocardial tissue was detected by Elisa.The protein levels ofβ1-AR,PI3KCG and P-Akt in myocardial tissue were detected by Western Blot,and the mRNA levels ofβ1-AR and PI3KCG in myocardial tissue were detected by Real-time PCR.Results Compared with blank group,the cardiac function of model group was decreased,the expressions of BNP,β1-AR andβ1-ARmRNA were significantly increased,and the expressions of PI3KCG,P-Akt and PI3KCGmRNA were significantly decreased.Compared with model group,the expressions of BNP,β1-AR andβ1-ARmRNA in all treatment groups were significantly decreased,while the expressions of PI3KCG and PI3KCGmRNA were significantly increased.The expression of P-Akt in medium dose,high dose and metoprolol was significantly increased.Conclusion Xiaoyin Huayu Prescription may inhibit cell apoptosis,reduce cell Ca 2+concentration,improve cardiac function and delay ventricular remodeling throughβ-AR signaling pathway.
