对叶百部组培快繁技术体系的优化

摘  要:目的筛选获得一套便捷高效的对叶百部组培快繁技术体系以应用于生产。方法以带芽茎段为试验材料,优化了外植体的灭菌方法,不同植物激素种类及其质量浓度对丛生芽诱导、增殖、生根等环节的影响,并对无菌苗进行了炼苗移栽。结果以2%次氯酸钠溶液处理外植体12min效果最佳,萌芽率77.77%,污染率5.57%,褐化率16.66%;MS+6-BA 2.5mg/L+KT 0.2mg/L+IAA 1.0mg/L为初代诱导的最佳培养基,诱导出的丛生芽数量较多,且叶绿、植株长势良好;MS+6-BA 2.0mg/L+KT 1.5mg/L+IAA 0.2mg/L为最佳的继代增殖培养基,增殖系数为5.3,显著高于其它处理,且丛生芽的长势良好,数量多,叶片大;不定芽生根最适的培养基为1/4MS+6-BA 1.0mg/L+NAA 0.5mg/L+活性炭500mg/L,生根率可达77%,显著高于其它处理;以泥炭土+蛭石为试管苗移栽的最佳基质,移栽30 d后成活率为100%。结论研究结果成功构建了对叶百部的组培快繁技术体系,符合工厂化育苗的要求,可为对叶百部健康种苗的产业化生产提供技术支持。Objective To screen and obtain a convenient and efficient system for the rapid propagation of Stemona tuberosa Lour.for application in production.Methods Using stem segments with buds as test materials,we optimized the sterilization method of explants,the effects of different plant hormone types and their mass concentrations on the induction,proliferation and rooting of clump buds,and carried out refining seedlings for transplanting.Results The best resultls were obtained by treating the explants with 2%sodium hypochlorite solution for 12 min,with 77.77%budding rate,5.57%contamination rate and 16.66%browning rate;MS+6-BA 2.5mg/L+KT 0.2mg/L+IAA 1.0mg/L was the best medium for primary induction,which induced a high number of clumped shoots with green leaves and good plant growth;MS+6 BA2.0mg/L+KT1.5mg/L+IAA 0.2mg/L was the best medium for successional proliferation,with a proliferation cofficient of 5.3,significantly higher than other treat-ments,and the clumped shoots grew well,with a large number of leaves and large leaves;the most suitable medium for rooting of adventitious shoots was 1/4MS+6-BA 1.0mg/L+NAA 0.5mg/L+activated charcoal 500mg/L.The rooting rate was 77%,which was significantly higher than other treatments.Conclusion The results of the study have sessfully established a fast breeding technology system for Stemona tuberosa Lour.which meets the requirements of factory nursery and can provide tech-nical support for the industrial production of healthy seedlings of Stermona tuberosa Lour.
