摘 要:旨在研究体外培养中添加罗格列酮(Rosiglitazone,RSG)对小鼠卵母细胞体外成熟(IVM)的影响。收集4~6周龄雌性小鼠卵母细胞,随机分为对照组和20μmol/L RSG组。在饱和湿度,38.5℃、5%CO_(2)培养12 h,统计卵母细胞第一极体排出率。使用DCFH-DA检测卵母细胞内活性氧(ROS);采用CMF2HC检测卵母细胞内谷胱甘肽(GSH)水平;采用JC-1检测卵母细胞内线粒体膜电位;采用实时荧光定量PCR测定抗氧化相关基因的表达水平。结果显示,RSG组卵母细胞第一极体排出率相较于对照组显著提高(P<0.05);与对照组相比,RSG组ROS水平明显降低,GSH和线粒体膜电位水平显著升高(P<0.05)。本研究还发现,RSG组卵母细胞中抗氧化相关基因GPX-3、CAT和SOD-2的mRNA转录水平均显著提高(P<0.05)。结果表明,在体外成熟过程中添加RSG可以提高小鼠卵母细胞成熟率,降低细胞内ROS堆积,提高细胞内GSH水平、卵母细胞线粒体功能和抗氧化基因表达水平,缓解氧化应激造成的损伤,提高卵母细胞体外成熟质量。The aim of this research was to study the effect of rosiglitazone(RSG)on in vitro maturation(IVM)of mouse oocytes.Oocytes from 4 to 6 weeks old female mice were collected and randomly divided into the control group and the 20μmol/L RSG treatment group.After incubating at 38.5℃ for 12 h at 5%CO_(2) and saturated humidity,the first pole exclusion rate was calculated.DCFH-DA was used to detect reactive oxygen species(ROS)in oocytes.Glutathione(GSH)level in oocytes was detected by CMF2HC.The mitochondrial membrane potential in oocytes was measured by JC-1.The expression levels of antioxidant related genes were determined by real-time fluorescence quantitative PCR.The results showed that the first pole exclusion rate of oocytes in RSG treatment group was significantly higher than that in control group(P<0.05).Compared with the control group,ROS levels in RSG group were significantly decreased,GSH and mitochondrial membrane potential levels were significantly increased(P<0.05).Compared with the control group,the mRNA transcription levels of antioxidant related genes GPX-3,CAT and SOD-2 in oocytes in RSG group were significantly increased(P<0.05).The results showed that the addition of RSG in the process of in vitro maturation can improve mouse oocyte maturation rate,reduce the accumulation of intracellular ROS,improve the level of intracellular GSH,improve oocytes mitochondrial function,improve the level of antioxidant gene expression,relieve the damage of oxidative stress,improve the quality of in vitro maturation of oocytes.
