摘  要:旨在研究低氧环境下不同浓度菊苣酸(chicoric acid,CA)对SD大鼠心肌组织低氧适应性的影响。选取60只6~8周龄、体质量为(192.00±7.35)g的健康雄性SD大鼠(P<0.05),随机分为对照组、不同浓度CA组(10,20,40 mg/kg),每组15只,连续灌胃49 d。灌胃50 d时采集SD大鼠心脏,并测定各组大鼠心肌组织活性氧(ROS)含量,抗氧化指标超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)、过氧化物酶(CAT)活性,线粒体功能相关指标线粒体呼吸链复合体Ⅰ、Ⅲ活性,能量代谢相关指标Na^(+)-K^(+)-ATP酶、Ca^(2+)-ATP酶活性及低氧适应基因缺氧诱导因子1α(HIF-1α)、促红细胞生成素(EPO)、血管内皮生长因子(VEGF)的mRNA及蛋白表达情况。结果显示:与对照组相比,不同浓度CA组显著降低ROS活性(P<0.05);显著降低MDA含量(P<0.05),显著提高SOD、GSH-Px、CAT活性(P<0.05);显著提高线粒体呼吸链复合体Ⅰ、Ⅲ活性(P<0.05);显著提高Na^(+)-K^(+)-ATP酶、Ca^(2+)-ATP酶活性(P<0.05);显著提高HIF-1α、EPO、VEGF的mRNA及蛋白表达(P<0.05)。各试验组间结果显示,40 mg/kg组显著降低ROS活性(P<0.05),显著降低MDA含量(P<0.05),显著提高SOD活性(P<0.05),极显著提高GSH-Px、CAT活性(P<0.01);20,40 mg/kg组极显著提高呼吸链复合体Ⅲ活性(P<0.01);40 mg/kg组极显著提高Na^(+)K^(+)-ATP酶活性(P<0.01),显著提高Ca^(2+)-ATP酶活性(P<0.05);40 mg/kg组极显著促进HIF-1α、EPO的mRNA表达(P<0.01),显著促进HIF-1α、EPO的蛋白表达(P<0.05)。结果表明,CA可通过抑制SD大鼠心肌组织ROS活性,提高其抗氧化能力、线粒体功能、能量代谢水平及低氧适应能力,增强SD大鼠心肌组织低氧适应性,且以40 mg/kg组作用最佳。This experiment was conducted to investigate the effect of different concentrations of cichoric acid(CA)on hypoxic adaptability of myocardial tissue in SD rats under hypoxic environment.Sixty healthy male SD rats aged 6 to 8 weeks(192.00±7.35)g were selected,SD rats were randomly divided into the control group,different concentrations of CA groups including 10 mg/kg group,20 mg/kg group,40 mg/kg group,15 mg/kg group,continuously feeding for 49 d.Feeding of the 50th day,the hearts of SD rats were collected,the content of reactive oxygen species(ROS),antioxidant indexes superoxide dismutase(SOD),malondialdehyde(MDA),glutathione peroxidase(GSH-Px),peroxidase(CAT),mitochondrial function related indexes,mitochondrial respiratory chain complexⅠandⅢactivities,energy metabolism related indexes Na^(+)-K^(+)-ATPase,Ca^(2+)-ATPase activity and expression of hypoxia adaptation gene hypoxia inducible factor 1α(HIF-1α),erythropoietin(EPO)and vascular endothelial growth factor(VEGF)were measured.The results showed that compared with the control group,the treatment of different concentrations of CA significantly reduced ROS activity(P<0.05),significantly reduced the content of antioxidant index MDA(P<0.05),increased the activities of SOD,GSH-Px and CAT(P<0.05),and significantly increased the activities of mitochondrial respiratory chain complexⅠandⅢ(P<0.05),significantly increase the activities of Na^(+)-K^(+)-ATPase and Ca^(2+)-ATPase(P<0.05),significantly increase the mRNA and protein expression of HIF-1α,EPO and VEGF(P<0.05).The results between test groups showed that,in 40mg/kg group,the activity of ROS was significantly decreased(P<0.05),the content of MDA was significantly reduced(P<0.05),and the activity of SOD was significantly increased(P<0.05),the activities of GSH-Px and CAT were extremely significant increased(P<0.01).In 20mg/kg and 40mg/kg groups,the activity of respiratory chain complexⅢwas extremely significant increased(P<0.01),in 40mg/kg group,the activity of Na^(+)-K^(+)-ATPase was extremely signi
