摘  要:该研究通过搅拌加热的方法首次成功制备了以腺嘌呤为有机配体、金属钛为核心的生物型金属有机骨架(MOF),Ti-MOF-79,并以槲皮素(1)为药物模型对Ti-MOF-79的载药、释药性能进行研究。通过扫描电镜(SEM)、X射线衍射(XRD)、差示扫描量热(DSC)、X射线光电子能谱(XPS)、红外光谱(IR)、紫外光谱(UV)和细胞毒性试验等方法对该材料和载药复合物进行表征,并计算了载药量及累积释放率。结果表明Ti-MOF-79是一种规则的圆柱体形晶体,具有多孔结构,载药量为(28.83±8.35)%,在体外具有一定的缓释作用,且无明显的细胞毒性,提示MOF有望成为新型载体材料。The biological metal-organic framework(MOF)Ti-MOF-79 with adenine as an organic ligand and Ti as a metal core was synthesized under hydrothermal conditions.Quercetin(1)was used as a drug model to investigate the drug loading and release performance of Ti-MOF-79.The material and the drug-loaded complexes were characterized through transmission electron microscopy(SEM),X-ray diffractometry(XRD),differential scanning calorimetry(DSC),X-ray photoelectron spectroscopy(XPS),infrared spectroscopy(IR),ultraviolet spectroscopy(UV)and cytotoxicity test,and the drug loading and cumulative release rates were calculated.The results showed that Ti-MOF-79 was a regular long cylindrical crystal with porous structure.The results of in vitro experiments showed that the complexes with drug loading of(28.83±8.35)%had a sustained-release profile and had no obvious cytotoxicity,which indicated that MOF could become a novel carrier material.
