摘 要:O-甲基转移酶(O-methyltransferase, OMT)是植物中调控次生代谢产物生物合成的一类关键酶,主要功能是催化酚类物质发生氧甲基化反应。本研究在白木香(Aquilaria sinensis)转录组中筛选到一个OMT编码基因AsOMT1,该基因与2-(2-苯乙基)色酮合成关键酶基因(PECPS)表达高度正相关。AsOMT1基因全长1 456 bp,包含一个1 074 bp的完整开放阅读框,编码357个氨基酸;氨基酸序列比对和系统进化聚类分析发现该As OMT1属于咖啡酸O-甲基转移酶亚家族。AsOMT1基因启动子区存在多个激素和胁迫应答元件;实时荧光定量PCR结果显示,伤害处理和乙烯处理均能显著上调AsOMT1的表达,推测其可能参与了沉香2-(2-苯乙基)色酮的生物合成。Plant O-methyltransferases(OMT) is one of the key enzymes in the regulation of secondary metabolites biosynthesis, which methylate the oxygen atom of phenolic compounds. AsOMT1 was screened by analyzing the transcriptome of Aquilaria sinensis, the expression of which is positively correlated with diarylpentanoid-producing polyketide synthase gene(PECPS). The full length of AsOMT1 is 1 456 bp with a 1 074 bp opening reading frame(ORF) and encoding a 375 amino acids protein. The amino acid sequence alignment results and phylogenetic analysis showed that AsOMT1 belongs to coffee acid O-methyltransferase types. Several responsive elements in response to hormones and stress are found in the promoter region of AsOMT1. Real-time PCR results showed that expression of AsOMT1 was significantly increased by wounding and ethephon treatment. These results indicated that AsOMT1 may be involved in the biosynthesis of 2-(2-phenethyl)chromone in agarwood.
