摘 要:目的鉴定十二味参芍益脑合剂化学成分,进行大鼠血清药物化学研究。方法采用Acquity UPLC HSS T3色谱柱(150 mm×2.1 mm,1.8μm),以水(含0.1%甲酸和0.1%甲酸铵)-乙腈为流动相进行梯度洗脱,电喷雾离子源正/负离子扫描模式采集质谱数据。大鼠灌胃10倍临床等效剂量,探究十二味参芍益脑合剂入血、入脑物质轮廓以及入血的代谢物。结果从十二味参芍益脑合剂中共鉴定出24个化学成分,其中芹菜素、黄芩素和甘草素等14个物质经过对照品确证。血清中检测到16个原型成分和11个代谢物,在脑组织中鉴定得到11个原型成分和1个代谢物。结论解析了十二味参芍益脑合剂的化学组成、制剂移行入血和入脑成分及其代谢物,可为该制剂的质量控制和药效物质基础的进一步研究提供实验依据。Objectivee To qualitatively analyze the chemical compositions of Shierwei Shenshao Yinao Mixture(SSYM)and the serum pharmaco-chemistry in rats by LC-MS technique.Methods The analysis was performed on Acquity UPLC HSS T3 column(150 mm×2.1 mm,1.8μm)with mobile phase of acetonitrile-0.1%formic acid and 0.1%ammonium formate water solution for gradient elution,the data of samples were collected in positive and negative ion modes.The serum pharmaco-chemistry analysis method was used to identify the migrating components in rat serum after intragastric administration of the SSYM.Results A total of 24 chemical components were identified from the SSYM,14 compounds including apigenin,baicalein and liquiritigenin etc were confirmed by reference standard.16 prototype components and 11 metabolites were found in serum and 1l prototype components and 1 metabolite were found in brain.Conclusion The chemical compositions and the migrating components in rat serum and brain of SSYM are illustrated,which is significant to the quality control and further study of material basis of efficacy of SSYM.
