摘 要:目的研究补阳还五汤对一氧化碳中毒迟发性脑病(DEACMP)大鼠氧化损伤的保护机制。方法改良式间隔腹腔注射法制作DEACMP大鼠模型,随机分为模型组(n=30)、补阳还五汤组(n=30)和空白组。HE染色检测病理变化,Tunel法检测细胞凋亡情况,生化实验检测血液中MDA、8-OHdG水平,qPCR检测HO-1、CAT、SOD、GST mRNA表达,Western Blotting检测Nrf2表达,coIP检测Nrf2与c-Maf的相互作用。结果补阳还五汤可改善DEACMP模型大鼠海马组织损伤,减少浸润的炎症细胞、组织水肿和细胞凋亡,降低DEACMP模型大鼠血液中MDA和8-OHdG含量,提高DEACMP模型海马组织中HO-1、CAT、SOD、GST mRNA表达以及Nrf2和Nrf2磷酸化水平,增加DEACMP模型大鼠海马中Nrf2与c-Maf蛋白的相互作用量。结论补阳还五汤可更进一步激活海马组织中Nrf2/ARE信号通路,保护海马组织免遭急性CO中毒所致的氧化应激损伤,预防DEACMP。Objective To study the protective mechanism of Buyang Huanwu decoction on oxidative damage in rats with delayed encephalopathy after carbon monoxide poisoning(DEACMP).Methods The DEACMP rat model was established by modified intraperitoneal injection method,they were randomly divided into DEACMP group(n=30),Buyang Huanwu decoction group(n=30)and control group.HE staining was used to detect pathological changes,tunel staining was used to detect apoptosis,the levels of MDA and 8-OHdG in blood were detected by biochemical test,the mRNA expressions of HO-1,CAT,SOD and GST were detected by qPCR,the expression of Nrf2 was detected by Western Blotting;CoIP detects the interaction between Nrf2 and c-Maf.Results Buyang Huanwu decoction could improve the damage of hippocampus,reduce the infiltration of inflammatory cells,tissue edema and apoptosis in DEACMP model rats,reduced the content of MDA and 8-OHdG in blood of DEACMP model rats,increased the expression of HO-1,CAT,SOD,GST mRNA and Nrf2 and Nrf2 phosphorylation level in hippocampus of DEACMP rats,and increased the interaction between Nrf2 and c-Maf protein in hippocampus of DEACMP rats.Conclusion Buyang Huanwu decoction can further activate Nrf2/ARE signal pathway in hippocampus,protect hippocampus from oxidative stress injury caused by acute CO poisoning,and prevent DEACMP.
