摘  要:为明确草本油料、木本油料与非油料作物内δ8-FAD2基因密码子的使用偏好性及其影响因素的差异,为进一步通过基因调控改良油料作物品质提供研究基础,从GenBank数据库选取15种不同类型植物的δ8-FAD2完整基因序列,对其密码子使用偏好性进行分析;并使用CodonW软件分析序列,得出A3、T3、C3、G3、GC3、有效密码子数(ENC)、相对同义密码子使用度(RSCU)等重要参数;通过相关指数分析、多种参照体系的最优密码子确定、ENC-plot绘图分析、PR2-plot分析及系统关系分析对比15种植物的基因序列。结果表明,15种植物δ8-FAD2基因密码子使用偏好性均较弱且偏好使用A或T结尾;δ8-FAD2基因共有17个优势密码子,最优密码子多达21个;不同植物δ8-FAD2基因的最优密码子有所差异。通过ENC-plot和PR2-plot分析得出,油料作物的δ8-FAD2基因密码子使用偏好性要强于非油料作物,木本植物间其密码子偏好性的受压力情况基本类似,而草本植物间各不相同。分别基于RSCU值和完整基因序列进行聚类分析,发现物种间的亲缘关系并不是密码子使用偏好性的决定性因素。To clarify the differences in the codon usage bias ofδ8-FAD2 gene and the influencing factors between herbal oil crops,woody oil crops,and non-oil crops,provide the research basis for further improving the quality of oil crops through gene regulation,in this study,the complete gene sequences ofδ8-FAD2 gene of 15 different types of plants were selected from GenBank database and their codon usage bias was analyzed.CodonW software was used to analyze the sequences to obtain important parameters such as A3,T3,C3,G3,GC3,effective codon number(ENC),and relative synonymous codon usage(RSCU).Correlation index analysis,optimal codon determination of multiple reference systems,ENC-plot analysis,PR2-plot analysis,and systematic relationship analysis were used to compare the gene sequences of 15 different plants.The results showed that theδ8-FAD2 codon usage bias of these 15 plant species was weak,and they preferred ending with A or T;theδ8-FAD2 gene had 17 dominant codons and 21 optimal codons;the optimal codon ofδ8-FAD2 gene in different plants was different.According to the analysis of ENC-plot and PR2-plot,the codon bias ofδ8-FAD2 gene in oil crops was stronger than that in non-oil crops,and the codon bias pressure was similar among woody plants,but different among herbal plants.Cluster analysis based on RSCU value and complete gene sequence showed that the genetic relationship between species was not the decisive factor for codon usage bias.
